Tissue Culture / Cloning Question

Was wondering, I want to get into cloning / tissue cultures. Question is, can I use 3rd generation G2G shrooms? Is tissue culture similar to g2g transfers, regarding generations? Is a  tissue culture from a shroom of a 3rd generation ( 2nd flush) G2G transfer weaker, and does it limit future generations g2g transfers?  Then can do multiple G2G transfers after master jar created? I believe 5 G2G transfers is kind of the max, without dedgredation. Is the tissue from a 3rd generation shroom  from 3rd grain transfer shroom, considered 1st generation or 4th? Hope this makes sense,lol.

My normal procedure- 7 qts of Grain,6 for spawn, 1-for G2G.repeat as tubs are needed.keep repeating until 5 G2G transfers take place, then either collect spores, or use agar.

Tissue culture rye jar to 7 jars= 1 generation,no true master jar, as you can only use them once after being exposed to create additional jars.

Reason being, have 4 types of cubes in ptheir 3rd generation tubs ( 3rd G2G spawn), however I made
Agar plates and pint jars for LC at 2nd transfer, but all from Ms Syringe.strains saved, but want to clone. Have no firt/ original harvests to work from.

If u clone mycelium u dont know what ur cloning. Grow it out and pick a few mushrooms ud like to replicate. Big ones or clusters or mutants. Whatever ur into.

And dont think about generations too much. Technically its still the first gen cuz u didnt start back from spores.

Ok, so a shroom from a 3rd generation tub, is considered new myc? I'm taking tissue from shrooms, not raw myc

You will be fine to take clones of those mushrooms but I have heard that you don't want to clone a clone. I've read it "waters" down the genes too much. So if u grow out a clone tub u can take prints but u wouldn't want to take a clone from it.

In you case your taking the tissue from a mushroom that has never been cloned so you are fine.

Won't clone a clone, but if I can get 3-5 generations of G2G transfers from a cloned master jar. Also would make agar from original cline grain jar to store. Don't want to be hassled to take tissue cultures if I can only  get 1 additional G2G transfer, from a 3rd generation tub. Trying to put it in lehmans terms,lol

Oh now I see what your saying, sorry. Yes it would be considered new myc since you pulled from a tissue sample. You could grain to grain that master 3-5 times.

Guess I could have just asked, is tissue from a shroom, the same age as a 3rd generation G2G jar / spawn used to take tissue  or is it considered new, original, like original inoculation.

Already made 14 jars, and 2 tubs of sub, and 7 jars of g2g not even lunch yet,lol.

Watering down genes?

Doesn't make much sense..

A problem I can see with it is you may to be growing too much in the direction of senescence.
The correcter way afaik would be to isolate a cloned piece of tissue on agar, to make an isolate. Then you should have a genetically pure culture and further cloning would be pointless (and all pointless growing gives you more senescence ultimately) if you did the isolation right and that's the case.

So yes it is also true that with more G2G transfers you also age your culture more and more and it is less ideal than cloning a much younger piece of tissue (age calculated by how much growth there was from the original spores). Whether it is actually problematic depends on the age and whether you approach senescence (old age).

I guess just do it if you have no choice: no younger myc/fruits. But eventually you may have to go through spores again and start a new generation if you don't get the vigor you expect.

^^ So shroom tissue is not new  / original myc? It's the age of the original spore germination?
