Nothing much more infuriating than shaking a jar to the brink of self-inflicted hemorrhage, only to put it down to find the wedge completely unperturbed and firmly fixed in its spot on the underside of the lid.

I'm trying dollar store dishes with the green lid for the first.  The lids are cheap and a few broke when drilling holes.  But the have a diameter similar to a standard petri so that's nice.



Agar for no pours can just be mixed with hot water, poured, and then PC'd in the containers... hence "no pour " bc you're pouring before sterilization ... It's not necessary to cook it with no pours,  the pc cycle will do the cooking and sterilization in one fail swoop... let them cool inside the pc over night... holes are easier to drill in the side of the container itself near the lid IME

I do this but I wait 5 days before shaking.

Why wait to shake? You've already disturbed the culture, might as well spread it and increase inoculation points... Just curious the reasoning behind it

Letting anything else that may of fallen in germinate so post shake you can tell if you got some mold or whatever in by accident.

Can also check recovery time of the grains that got colonized in the last few days. Gives an idea if it's bacterial or not.

Thanks for the tips, for some reason I thought you had to boil to get the agar and LME to mix well.

I used my SAGARI dishes this morning.  Transferred T4 F+ to an Agar dish and to a SAGARI dish.  Will run both of them against each other on grain once they grow out.

Soft agar shake will win Everytime.

You need to boil the water to dissolve the agar. Dissolving sugar in water is easier if it's hot. Usually I'll heat add agar dissolve it and then add the sugar last.

Just mix with hot water and let the PC cook that shit... or boil an excess amount and do your measuring when it's hot... But hot tap water will disolve it... and the idea is to get fully colonized spawn  prior to germination of contaminant spores, not to wait on it to catch up... I'm so confused lmao... Work on (or trust) your sterile technique, identifying a clean culture, and proper grain prep, and you'll be fine ...

I know I'll be fine. But contamination does happen even if we think our tech is flawless.

I'd rather weed the bad seeds out before I commit 5lbs of grain and a bag. Easier to dump the jar imo.


Also I'll add that mold spores can germinate in 24 hours. And I know it takes longer than 24 hours for a shaken to shit agar puck to colonize everything. I'd just rather be on the safe side.

Id like to try this too, but Im stuck at the lid for the container.  I need something that allows for gas exchange yet is still conducive to shaking with a liquid inside.

Parafilm?  Would that work as a cover?

Like replacing the petri lid with it? I've never used it, but I'd imagine it'd make a mess... I just shake the grain jars after dropping :shrug:

I think the write-up would be better with more pictures of the actual Tek showing the benefits. Right now it's more like a rushed write-up of how to make soft agar, no offense.

I'd love pics of: the myc growing on the plates, definitely jars post-inoculation (before shaking),and then after the initial shake to show distribution. I think the texture and firmness of the agar is a key component and not everyone's ratios will produce what you probably consider optimal so a visual guide might help.

I'd also personally love side-by-sides of say, grain with your softer agar and a regular puck to demonstrate the speed up of colonization times.

I've never gotten 100% colonization in 4 days with anything and I primarily use liquids + G2G. But being fair, like 90% of my growing is done with a very small range of genetics and I haven't spent time looking for quicker cultures. Need to quit being lazy. Edit: This isn't me doubting you, just commenting that my shit always seems slow in comparison to others.

Curious, you ever tried Munch's BLI tek? It's essentially soft agar tossed into sterilized water and hand shaken. It's a good alternative to traditional LI and LC if you're not a fan of liquids.

I'm also curious if you've tried this with Pasty Plates? I think the grid pattern is easier and faster to dislodge without the scalpel scooping them. That's how I'd done them, just invert the bottom to loosen the puck (like a Tiger Drop), cut the grid, and dump.

Also, why do you unscrew the cap to your agar bottle and then start cutting plates from the sleeve? Why wouldn't you have everything setup and ready before?

I'm working on pictures, but  won't have anything substantial until after thanksgiving traffic... totally open to collaborate and include any photos if you'd like to give it a shot? 4 days was once, and I use pints, 5-7 days is typical... not tried LI or LC of any sort: really stumbled on this initially making the mistake of using cold water and filtering prior to heating and presumably lost a good bit of the agar powder... I like it, and it avoids the extra steps... I don't care for no-pours, the containers were a PITA for me, so not tried this with them... have what setup and ready before? I don't open my sterile sleeve, or remove the tyvek from my agar until I'm pouring, everything else is set up... and then I go burn one... and if that's the case,  I'd imagine parafilm not doing so well in a PC :shrug:

Oh, didn't realize you were using pints. Might want to edit that in there. You should check out Munch's BLI and give it a shot. It's literally soft agar dropped in a bottle, shaken, and voila.

Parafilm melts in a PC. I was saying I thought Vroomer was talking about shaking the agar bottle, not the petri dish (hopefully).

Sorry, I didn't realize you stuff Tyvek in between the cap and media, so this part confused me: "After removing the cap, I cut open my sleeve of trays with my scalpel"

Why did you put a hole in your cap? I just realized. You don't need a filter for the gl45 caps.
