Problems with Agar Innoculation?

I have a question I'm hoping someone can answer for me.



Someone I know steralized an agar solution two+ days ago. The agar, I hear, is top notch (from fungi perfecti). This person poured in the agar, let it cool, then used an innoculation loop to scrape off some spores into the dishes (from a print). Its been over two days now and there is no sign of growth on any of the petri dishes. Just the spores sitting there. The room temperature is about 79 degreees farenheight or so. The only other thing he can think of is the spores are about 2 months old, but he didn't think that would have been the cause of it (good spores, I assume, too, from Hawk Eye).



Now, this person would like to know, is he doing anything wrong? There is no growth yet, and as he understand it, most dishes fill out in about 7 days, so he figured he should at least have some sign of growth. He hasn't done this technique before and would like to know iis there any special way to innoculate agar? Should it be done when it is still liquid/cooling, or after it has solidified? Is this lack of growth normal, and he should hang tight, or should he run another batch?

Give it another few days. It can take up to 30 days for spores to germinate, though my friend has never had to wait more than two weeks (on some very old spores).

My friend just germinated some spores (b+ and koh) from The Hakws Eye on agar from fungi perfecti a few weeks ago and they did fine. It took about four or five days before she started to see growth on the dish.

Yeah I've got spores that are 3 years old and they still germinate on agar with enough time. It does take like 2 weeks sometimes though. just give it some time
