I realize this is a stupid question but I still need an answer, what is the best way to get the agar off the blade when transferring to the new plate? I always have the problem of the agar getting stuck to the blade and not coming off and I don't know how to avoid touching it just to get it off. The only thing that seems to work from time to time is cutting bigger pieces of agar which gives me bigger sections of mycelium which are likely to be different sectors. Its easier to lay them down that way though so I'm not sure how to avoid it and hope for the best.

A few things can help:
  • Softer agar will stick to the blade more. If you make a softer agar recipe, add a little more powder (don't do this if you aren't using a softer agar recipe)
  • If your blade is still too hot it can kinda melt the agar rather than cut it and cause it to stick. Make sure it's cool.
  • Turn the transfer myc side down then put it on the new plate. The myc will kinda stick to the surface of the new plate. This also can help the transfer start to grow out a bit quicker. Don't do this if you have to sacrifice good sterile technique to make it work. 
  • scoop your agar up on your blade so it easily comes off. Also, you can just stab it just enough to pick it up and it will come off the blade easily


It all just comes to practice. We've all been there. Still happens sometimes. It's annoying as fuck

Hell yeah thanks man!! Also I was wondering if its ok to use the agar that I'm not using to isolate from, if I'm isolating from plate A and plate B is not a monoculture but no contams will I have any issues putting it into grains?

As long as its clean, there shouldn't be any problems. If it's from MS you may get a more uneven canopy but that's not a problem at all. Go for it
