First go at syringe making... How's this sound?

This is with a nice dark Burma about 1.5" across. Thanks just_me and uncle :thumbup:

I figure I might as well do up the whole print and that I should be able to get five x 12ml.  I plan to boil DH2O in a saucepan with a little KY jelly to limit clumping.  I'll then pull it into a new 60ml sterile-packed syringe.  This will be recapped and placed in a ziplock bag sprayed with lysol and allowed to cool. In a still-air box I'll transfer the spores to a small glass dish, wiped with alcohol shortly before, along with the 60ml of DH2O. I plan to suck it back in the 60ml syringe and squirt it back out to get a good mix. I then fill five new 12ml syringes as 5x12=60.

How does this sound, people?

I'm also wondering if I can rinse the spores off the foil with the water from the 60ml or if I should scrape them off.

you dont want to use the whole print
a tiny corner is more that enough to make a syringe

that print is not 100% sterile and you would be mixing all the contams
pu if you use the whole thing

possibly contaminating every syringe you make

I also made my first endeavor into syringe making. I followed Magash's tek as well. I had a few questions about it myself here.
If you follow the tek and use a glovebox there is nothing to it. Personally I like dark needles, and from 1 3" diameter print I did 3 needles. I probably could have done 20 or more with this print, and will adjust accordingly next time.

On the subject of dark needles tho, I had a Tex syringe that was as dark as night and from that my 1/2 pint PF inoculated jars were colonized %100 in 6 days.

There is something to be said about making your own needles. ;D

inside your glovebox put a sterile shotglass your print, and a couple of syringes already filled with sterile water and whatever tool you use for scraping your spores. 

scrape about a 1/8" square off into the glass then squirt your syringe water in the glass and swirl with needle still in the water and fill your syringe back up.  repeat as needed. 

pending how long ago/fresh your print is you can use right away or let sit 4 a day to rehydrate your spores.

I hadn't considered that using all the spores at once might increase contamination odds, although that now seems obvious. No way I need five stick worth right now. Just one is probably the way to go.

Do you use the syringe needle to scrape the spores?  I don't have an inoculation loupe an the needle will already be sterile.

I used the needle tip. From further investigation you can damage the spores from doing this, using the sharp needle tip to scrape them in the shot glass.

The best suggestion I have read; if you don't have an innocu loop, is to use a Japanese ear cleaner. It is long w/ soft round edges, they come in plastic and wood. I had one, but didn't read this until I already made the needles.

Using the tips turned out fine, just food for thought since you asked.

Using a the tip of a syringe is fine damaging a few spores wont really matter since there TONNES of them in a syringe even if you cant see lots of them(which is what you want)

I have to ask:
What the hell is a Japanese ear cleaner, and do they only clean Japanese ears?

its likely you will kill a few of them

but whats 1000 when you still have ten thousand
left in the syringe

you will be ok for now
but it will be well worth it to pick one up

Well, I did the deed.  I dusted this off out of the old posts:

It sounded silly at first, but the more I thought about it, the more I liked, and at least one TC is down with it.

Seemed to go well and the syringe looks good. We'll see how it works out.

Yeah, I've seen that type tek and it looks good, but I didn't make the print. It was on foil and I only made one stick. I don't think I've seen that exact thread though and it looks to have some good stuff. Thanks :thumbup:

1cc is used in a 1/2 pint and 1cc - 2cc is used in 1 quart jars so anywhere in between is good. :super:
