Antibac Agar, DIY!
    #1105063 - 12/02/02 09:12 PM (14 years, 3 months ago)

Joshua Says:
"The agar I am using now, I got through a trade from a fellow Shroomerite. Here's a recipe from The Mushroom Cultivator:
MEA

20 grams tan malt
2 grams yeast
20 grams agar

Add .10 grams of 60-80% pure gentamyacin sulfate to create an antibacterial mix."

But I got a question. Say someone had some agar and admixtures, and some 100mg trimethoprim pills. Trimethoprim is an antibiotic, I don't know what class its in.
Is it possible to make antibacterial agar with this? If so, how much antibiotic would be best?
And where might I find some gentamyacin sulfate? No links, just give me a category to look at and what its kept near/used for if it can be had in a store.
Thanks, y'all

The reason that gentamycin sulfate can be used is because it can withstand the temperature (121 degrees C) of pressure cooking. All other antibiotics that I know of will break down.
Why exactly do you want to use antibiotic agar? It's very rarely necessary. Look into using peroxidated agar, it's far easier cheaper and works like a dream.
You will not find gentamycin sulfate in any stores. Sporeworks sells it though

I want to use antibacterial agar to start spores to isolate the fungus what looks the strongest... and I dont have a flow hood or glove box.
I want antibac agar for starting spores, then I'd use the peroxide.
Sporeworks, eh? Thanks.
I love Tool.

You could just streak the spores and then re-isolate a clean sector onto peroxidated agar.
Same thing really.

Cheaper, too!
Call me stoopid, but you'll have to explain the streaking spores to me.  Would I throw some spores in some water?  I haven't handled spores outside of a syringe.  Will they stick to the wire on the streaking utensil?
Streaking spores.... naked spores?  Spore=seed, sorta.  Naked seed.  Gymnosperm.
Your screen name looks like an anagram, but if it is I can't figure it out.

Streaking is done with an innoculation loop of sorts.
You wipe the loop over some spores and they should stick to the loop.
Wipe those spores onto a particular area of the agar, preferably the center of the agar.
An innoculation loop can be a bent piece of wire or a professional loop.

You can germinate spores on peroxidated agar.
Just wet and sterilise some squares of cardboard, about 1cm x 1cm. The kind that's on the back of notepads. Smear some spores onto the bits of cardboard and stick them on peroxidated agar (2 per petri) and the spores will germinate on the cardboard, then the mycelium grows out over the agar.

Or you could use the oven.

i don't have an oven. also, over at mycotopia forums the mods/admins don't support the oven tek(not that that means a damn thing). they say the air flow is actually not conducive to sterility. there's a picture showing the air flow with the smoke from an incense stick. personally, i think you could do agar work in open air if you're careful. doesn't myc grow slower on treated agar? also, is there a bleach agar tek? it seems like something hippie3 would be working on.

The oven tek works rhizo.
It has been debated many times but I can tell you from my experiences that it really does work.
You really have the choice of using it or not.
I personally do soo much work that the oven is really the easiest and fastest option.
