Experiments in Storeing Cultures Under Mineral Oil
    #91524 - 01/03/00 09:55 PM (24 years, 8 months ago)

They were stored with and without mineral oil, for six years.
Mineral oil = produced sporeless fruitbodies (sterile).
Without = were extremely viable and produced fertile offspring.
I would suggest water layers as an alternative or maybe (10% glycerin 90% water) for the dung dwellers.

So you stored the mycelium under mineral water for 6 years and when the mycelium fruited it produced mushrooms without spores? I've heard that ingesting spores can cause upset stomach and things of that nature, could this possibly be a good thing?

Storing under mineral oil shows miajor signs of degeneration. Some of the effects were sporeless, irregular growth, and the inability to orient themselves correctly. This is a bad thing.

Moe Howard
Don,t store cultures under mineral oil.
The control group show les degradation and they were stored with NOTHING they sat in a frig for six years.
It only benifits those who have a cultures which they love and want to keep for long period of time.

HERE A FEW METHODS
Mineral oil storage; for mycelium.
Water storage; plug method,or water layer; for spores or mycelium.
Soil storage or compost; for mycelium.
Sillica gel storagr; only for spores.
Variations of the above using glycerin instead of water.
Freezing under giycerin; for spores and mycelium.
These are the most commonly used.

Hey Doc . . . parafin does work ! I don't do this myself, however, back in the dark ages ('60s), we DID use parafin for shroom/spore, long-term storage!

I must also thank Prof. Wilkenssen (I always suspected he was "sporing" more than shitakes)

I tryed to quick freeze mycelium using a CO2 quick release gun while the mycelium was in a container and sealed it and threw into a freezer
I hope that it wouldn't damage the tissue of the mycelium thats why I froze it in a 5second period with the gun but I prayed...and it worked somehow I dont know how but after thawing it was able to produce fruit of couse I have no control on this experiment (as in years) but hey if it worked maybe it'll last years to come as long as it is in a sorta air tite vaccum container... If you have any questions about this or comments please reply!!!

I believe that once the mycellium is frozen with the CO2 gun it can last for a very long time frozen. The damage in freezing occurs when the water in the mycellium reaches 32f and at this temp the water expands and bursts the cells. I am curious as to why the cells did not burst as the mycellium thawed and was exposed to a much longer period of 32f than the quick freeze. I think the praying must have done it.
The damage in the freezer is from drying so an airtight container is a must.
Maybe you will develope a strain of freeze tolerant Cubensis or Pan tropicalis to grow in northern gardens--just wishing.
