Agar Cloning vs Isolation

OK let me give you guys some background.  I just did my first grow on monotubs and it went great.  I started from multispore and I know its time to restart but I want to start with an isolation of something that is potent and fast growing.  I have seen Roger Rabbits videos and am a little confused on cloning vs isolation.  Lets say a monster mushroom grows and I want all of my mushrooms to be monsters just like it.  I can cut a piece of it put it on agar and use that to inoculate things and they will all be monsters?  Now what makes cloning different from isolation?  If I put a sliver of that monster mushroom on a petri dish, then can I isolate it to a monoculture and have something that is really potent, fast growing, and large?  Also on a side note I have a question about spore works petri dishes... http://sporeworks.com/store/product.php?productid=16247&cat=258&page=1 ; Do these petri dishes have 3 sections in each dish like in the roger rabbit videos?

Cloning is taking a tissue sample from a fruit.  Isolation is generating numerous strains from spores. With isolation, you want to fruit each strain to pick the best performers.

One thing on cloning that I just learned (RR Credit here) is that size is greatly dependent on correct moisture content and genetics is secondary. Cloning for growth rate and number of pins and probably other factors is what you want.

I guess one section petris, but didn't look that close behind the watermark... I have not found 3 or 4 sectioned petri dishes form sponsors, so I don't think it would be a problem to post this http://cynmar.com/item_detail.aspx?ItemCode=15023321 <-- mods, edit this if you want...

Cloning a large fruit doesn't neccessarily mean you'll get large fruits, especially on a first flush with a nice pinset. Usually the larger fruits will come during the later flushes when there aren't as many mushrooms competing for nutrients.

with isolation you will get consistant results. with cloning there are more than one substrain creating a single fruit. so a negative trait could show itself. also with isolation you shoot for the fastest rhizo growth each time you transfer. so you end up with really fast colonizing substrains to fruit and pick from. speed may be a gamble with cloning.
