liquid culture w/ sugared honey & what ratio?
    #6427784 -

i an extra spore syringe left and thought i'd try LC with honey. a few problems & questions i need to sort out before doing anything:

1. the honey has sugared. if i put the jar in a bowl full of hot water and the honey melts to a former self. can i use this or do i have to get unsugared honey?

2. the ratio of honey for 100ml of water.

just put a few drops of honey in, it doesnt have to be a whole lot. what i did was put a little bit of honey (maybe 1/20th of the amount of water i used) in a syringe and pressure cooked it. I pressure cooked the syringe in a glass jar with water in it. Once it was finished i put the mycelia in a jar with sterile water and shook it well. then you put the syringe in and draw up some water. Make sure you target the chunks of mycelia in the water for best results.

dunno about the honey being sugared but I'm guessing it would be fine.

for every 100ml of water add roughly 4ml of honey. 3-5% honey is fine but I wouldn't go much over or below that. 4% is optimal

it's where the honey starts to get hard/chunky. to me it resembles as if you mixed sugar into it

but yea, warm the honey up and if you have one, use an old syringe, take the needle off and measure it out that way. works great for me and the honey sucks up a lot easier after it's warmed

ok, warmed it up, injected 4cc's of warmed honey into 400ml of bottled water & boiled it a little for sterilization purposes just in case. added marble. in the syringe were spores clumped together or it was infected. i knew i could be sure so i'm just waiting and seeing where it goes. if it's infected the black sport should grow & spread, right? anyway when i move the marble around in there from the bottom some pieces float around that are whitish. now i want to make a jar out of it and see if it works: do i have to suck up those white pieces or is the water itself mixed with myc so much that anything from it will start colonizing?

Typically LCs are sterilized in a pressure cooker you did that right? You usually can't just boil the water and inoculate.

Often times spores will clump together in syringes you should shake before innoculating a jar.

LC contrams can be somewhat tricky to identify but nice white cloudy wisps are usually good sign. The black would probably be growing if it were something other than spores.

Best way to test a LC is inoculate some jars of grain/cakes. Should see growth in 72 hours or so.

If you used the fourth of the original honey LC recipe(witch is 4% honey 96% water) mycelium has eaten the food in the water much faster.

Incubate for few days and it will double it's size if the recipe isn't too weak. You'll be able to see mycelium and maybe perhaps something goo on the bottom. Mycelium is formed as a cloud and the other goo is just everywhere. YOu want to collect the mycelium only to your syringe and leave the goo behind.

I usually move the mycelium again to the second LC and let it double it's size before inoculating it to the grain.

I like to add few cc's 3%peroxide to the LC five minutes before collecting the mycelium to the syringe. This pulls the mycelium up with bubbles and seperates the goo from the mycelium pretty easily.

If you have 1 cup of water 9 or 10ml of honey is about 4%.
