Why do agar?

I understand that agar is a good growth medium for mycelium and is great for cloning, separating a strain from contams, and allows you to choose the most prolific mycelium.
What I don't understand if your not cloning, and are pretty confident your spores are contam free why do agar work? Does choosing the most prolific mycelium and transferring it to a grain have a lot more benefits than just inoculating grain with LC?

yes. You can use a couple spores to start one plate, then transfer several cubes of the mycelium to different jars. When your plate is almost out of mycelium, you can transfer another block to another plate to start the cycle again.

definitely.
when u are doing ms on agar, u can sector and isolate the best growing substrains from the dish and create a somewhat pure culture of it.
thereby eliminating slow growing, non rhizomorphic strains and just going for the fast and strong genetics u are aiming for.
if u inoculate using a ms syringe, u cannot chose as there will be millions of different substrains - lacking the specific characteristics of the one isolated sub. peace ohm

so once you isolate the substrain and all that good stuff.. and you get prints from your first flush, will the prints keep all the characteristics that you got from doing the agar to begin with or dose it start all over every time you do a MS inoculation??

why?? if the strain was isolated why dont it keep the same characteristics?? i know there are millions of spores in each cap but i thought that with agar you would be pulling out "1" strain or characteristics out of the million.. i think im just babbling.. pay no mind to me..

Yes the only way you can stick with those genetics is to store that culture. Apparently you can keep it active for a very long time if you periodically switch the nutrient base that you use for you agar recipe. Keeping cultures on the same nutrient base for a long period of time will lead to the mycelium developing more specific enzymes.

Look at spores as + and - . Hyphae fusion occurs between + and -. So when + finds - a whole new set of genetics is developed.

with a multispore inoculation you will get traits from the parent mushroom. the mutations will cancelout or be overrun with the more common spores. its like taking the average of all the spores.

also if you do agar a head of time you can have a lot fast colonization. Plus you can store them in a fridge for use later.

can agar cultures be done in on the bottom of a 1/2 pint jar? I don't have petri dishes.

one more question,
The Asian market I shop at has, believe it or not, a dried seaweed aisle. They have bags of agar. It looks like clear flakes. Is this the same thing used in agar recipes in the "FAQ's" section?

Agar is lots and lots of fun!
you can really get a good visual sense of what you're dealing with. I used agar to isolate a pure clean strain from a wild print of p. caerulipes... i've yet to do anything with it yet... but by god it was a blast!
