Potabello Mycillium on Agar

I took a print from a port a got at the store. Im trying to get the spores going on Sabaraunds (sp) Agar. On the few attempts that I try, I have gotten a blue-green mold colony overgrowth. I cant even seem to get it seperated out by dilution/streaking.

Any ideas/suggestions?

Clone the mushroom, it?s far easier.
Use peroxide agar, if you don?t have a flowhood.

I never tried cloning before... but it I should be able to pull it off.

When you say peroxide agar, do you mean that I should add peroxide to the PDA or MDA agar? Im not really sure what you mean here, as I just scanned the site that you refered me too. I'll give it a thorough read tomorrow.

An update on the project.

I bought a port at the grocery store. Took some tissue samples. They cultured nicely to agar! The tissue samples from the region of the cap and the gills grew the best. I generally got poor results from tissue samples removed from the stem.

Anyhow, I innoculated 3 quart jars of malted barley (a friend had it left over from a beer-making project). The culture took in only one of the jars. Never could get the other 2 to grow... even after re-innoculating.

Anyhow... Im about to case in the next day or so... any suggestions would be helpful. Im planning on straw and composted manure.
