agar problems

I am STILL trying to get the hang of agar.

5 days ago I inoculated 4 birdseed jars with Pink Oyster wedges. Since I suspected that the oyster plate used was contaminated, I put wedges onto 6 new peroxidated PDA plates as well.

Today I found that all the birdseed jars had nice growth, although two of them had the green. Of the 6 PDA plates, NONE are showing growth.

Would the proper amount of peroxide slow growth that much? Was PDA a poor choice for oysters? Or is it more likely that I just fucked up the recipe?  :confused:

I'm almost out of wedges... if I mess up again I'll lose my oysters.  :frown:

If youve moved to pda from another agar recipe, then it may take a bit to adjust to the new agar, as well as the peroxide.What temp do you have them at?
And do you think you could have over boiled the potatoes?As i did this once and it slowed everything down.

I really do not know what the previous aqgar recipe was, the plate was given to me by a friend. Incubaton fluctuates between 79F and 84F. If it's possible to over boil potatoes then I probably did it.  :crazy:

I'll give these plates another 5 days or so... and for my final attempt I'll us MEA. Thanks bro.

try dog food agar DFA 10grams dog food - 10 grams agar - 500 ml water. and poof!

You should wait more than 5 days, it takes some time to recover.
Pink oysters sometimes switch to "fruit mode" and start slowly to develop primordia on agar, even before the plate if fully colonized.

My Pink Oysters are safe.

After 5 more days of no growth, I mixed up a batch of MEA and tried again with the last of the wedges. Several plates are now growing nicely.  :laugh:

I must have REALLY messed up that PDA; not even the green will grow on it.  :grin:
