Eh... how?

Do you cut a wedge and drop it in some peroxidated honey water, or maybe just scrape the mycelium off a dish and add it to water? With or without peroxide?

I usually prepare a dextrose+water jar.
I simply drop an agar wedge in it.
Without any peroxide.
Performed in a glovebox, of course.

I have an eberbach now, but before I would take a blender base and unscrew it (the blade section, from the glass pitcher). Then I take a wedge and toss it in a sterile jar, in front of my hood. Add some boiled water (cool now) in front of hood to jar. Screw the bladed base on to the jar, pop it onto the motor, pulse a couple times. Open in front of hood, suck up solution into syringe. And that's that.

I dropped an agar wedge into peroxidated fructose/potato water. 3 out of the 4 have turned cloudy. Agrocybe Agerita turned cloudy within 36 hours. I dropped it carrying it into my bedroom and some water leaked out. That may have been why.

I have some agar colonizing in some half pint jars. When they are colonized I'll try filling them with peroxide/water and scraping the mycelium off into it. That should be enough for about 20 syringes.

I usaly scrape of the surface mycelium
from a petri, knock the scrapings into a jar
of dextrose water with peroxide and two pieces
of broken glass.Leave for a day fo so, shake well
and use.
I find its much slower to take of than spores though :-(

That is the way I have done it so far. But instead of using a syringe I would just pour it into the bags. I have had bad results this way, due to contams. So next time I will use a syringe for inoculation.

TMC also says to squirt water onto your petri, and scrape with your syringe, then suck it back up.

Then you have liquid innoculum. I've never done it this way though. I have heard many here who have, however.

Might be the easiest route.

That was the first route i took, but i found it hard to get any
mycelium into the syringe  :confused: so i moved over to this way,
now i have a flow hood though, i would do it in a less paranoid way, and
hopefully have more time to get some into the syringe.
