Hello! Do you pay attention to temperatures of the place where you store your pan cultures?

At what temperatures is the growth hindered?

I'd say optimal growth is around 24C +/- 2 degrees, somewhere in that range, anything over 28-30C and things are starting to get too warm,
more so for bulk grows where the substrate does produce additional heat. The colder it is the slower they grow. When I am getting ready for culting
season in spring and doing germination, LC revival and cleanup the room temperature is at around 14C. things are slow, like a bit slower than
regular cubes at normal speed, but that does suit me. Anything colder than that and I wouldn't really call it room temperature anymore.

Are you asking because you just want to keep the heating bill low or do you want to slow down growth on purpose?

Thanks for the responses!

I don't have heating in my house, sadly, and there are drafts coming through windows because the handles are broken.

When I run the dehydrator or the autoclave the temperature increases, but during winter it goes as low as 11°C.

I have built a fermentation chamber, which is a little fridge that was modified with a temperature controller and a heating cable to rise temperatures inside, so that should work well to eventually incubate cultures to keep them properly heated.

Right now the temperatures inside are around 17-18°C most of the time, a little lower during night time.

I am growing IN01 for the first time, these plates (60mm) are T1s on PDA GG, that much growth was achieved in 14 days (sigh).

And here is a close up of the leading edge, maybe it helps with understanding what is going on.

The myc is not aerial only on the edges, I thought it might be because of the low liveliness cause of the temperatures, or maybe it's a IN01 thing. But probably it's just me coping with the fact that I want it to work soo badly that I become avoidant of the possibility of contamination.
I did already transfer these plates (and also sent them yolo)

I thought to ask here since the IN01 official thread has less interactions.

I feel you, have a drafty house as well and don't do any culture work in winter. At the moment I'm in about the same temperature range like you and
it took 4 weeks for jars to colonize, I don't mind plates being slow but jars are a pit painful to watch creeping along.

The fridge is a good idea, maybe I build one as well if I ever get into beer brewing.

These plates don't look bad, photos are a little blurry though. If you can't get aerial myc then run them through a cabin, try island cleanup or putting on BRF puck.

Finally got spores (thank you) and germinated 2 WA plates, and 2 plates of .75% lme agar of both TTBVI and Komoto/BVI. I scraped the spores into the plate with a flame sterilized scalpel for 1/2, and used a sterile swab for the other 1/2 of the plates.

We will see what happens?

Just in time, pretty sure my “fluffy myc tranfers” involved more mold than Pan myc.

Thanks for the response, it puts my worries on ease knowing succesful growers like yourself work on similar conditions.

If you need any help with building the fermentation chamber hit me up, I have a farily cheap and simple setup.

I took 3 wedge transfers and one aerial myc transfer, after 3 days the wedges are all fluffy, but the aerial myc is not showing any movement.

Should have taken more aerial transfers-

Next transfer I'll do it on an island plate.

First two yeah maybe, it's always hard to tell with the lack of detail in the images, makes it look like one creamy blob.

The third one, lol......was thrown in the mix for entertainment, right?
:uhhnothanks:

I hope that one is not related to the first two.

It's from pan spore n t1 soooo it's newer and looks contam only relation to others is it's pan?

Dont judge lol I'm still getting used to agar sandwiches n I lost my scalpel for about 2 months.. don't ask what I been using to transfer with :laugh2:

OK, I won't be asking then , I was just about to suggest to take smaller transfers .
You could try using a syringe needle on a nail for taking transfers till you find the scalpel. It's all I use.

Yeah, the third plate is sporulating green mold.

:laugh2: it was a small transfer xd less than half the size of a grain of rice for example n was aerial myc I failed at getting the myc off n onto the cabin for the sandwich xd

Entirely my fault with failure to operate quickly smoothly n not having to scrape across the agar for like a minute back n forth with lid off 💀 yea I'm embarrassed about that one though I learned from it  I mighta evin dropped it once I was clumsy af with that tool I'm glad I found my scalpel today

I also tried charcoal agar with pans. I have some plated that look really perfect on regular agar and on charcoal with the same cultures they HATE it, the mycelium looks terrible on them they seem to just scatter everywhere.. I wont do it again

I didn't even pick up on the black color and you are absolutely right. Quite a few people had weird growth on agar when it was the latest trend
and a lot gave it a go. That culture in the 3rd pick is sporulating mold though.

Definitely agree,  I used activated charcoal few years back on my last culti runs and Pans did not like it at all.

Got some purple gel and seems to work decently for contrast

Whats purple gel? I used purple dye once and all the culture on it just died because it had red#5 dye which is anti fungal by nature

Got a better phone n forgot to upload better pic of third platexd I personally don't think it's got green at all though it entirely could be commingled with some kinds mold there's at least a plin forming in the center

100% agree they act weird on charcoal n might just use the plates I make instead of the charcoals for pans going forward

And here's my culture of kamodoxbvi the slower culture of the 2 I isolated though this one looks like mycelium not mold xd.

Think I needs transfer more or send it?

Ah, these are much better photos. The previous image of the charcoal plate was throwing me way off as it had a green hue and the shadows made it look
like there was granular green mold all over. This time it looks like there is a lot of knotting going on, I would keep this plate around and see how
it is developing, could well be that you are getting pins on that one. Probably nothing wrong with it and could take transfers as well.

The second plate does look alright, I'd take an aerial myc transfer (or more) from it to keep backups and send the rest. That way you start getting
at least a result (good or bad) instead of just guessing and next time around you will have a better feel for it based on that experience.

The other plates looks so uniform that I'd take em from all around.
