Castellani method - do I need to minimise the amount of nutrient agar in the vial to promote stasis?

I’ve been using Castellani method with 2ml vials but am a bit concerned that the amount of nutrition in my agar wedges dropped into the vial may be too high?

Am I overthinking it or should I be attempting to minimise the amount of nutrition that ends up in the vial?

I've done 0,5% and 2% agar to 2ml vials with RO water. Both have revived within 4 days after 20 months in stasis.

Thank you too man I'm happy to help, this question came just in time, it's only the last month I have tried reviving the cryo vials I did when I first started doing them, revived 4 so far and everyone came to life real quick.

I did a couple of extra vials from the same plate the first run so I'll try reviving the next in a couple of more years.

Did your agar solidify the water? Mine have basically swelled up to where they look like sslc with a drop or 2 of water on top.

I try to slice horizontally close to the surface so I just get the top, but that's also so I can fit more slices in the tube since they're only 5ML and I pack em tight.

My understanding is you also want to minimize air, so I fill the water to the brim too.

Not really, I can see that the wedge is smaller coming out then going in so I guess some agar made it to the water. I have been dumping out the water leaving the wedge behind in the vial before I get it out and it's pours like water, if that makes sense :lol:

Yeah makes total sense. I think I may have over packed mine and the agar swelled. Oh well as long as they revive, or at least my dried grains revive I'll be happy

So far 100% has revived, but not that old yet, and thanks for reminding me to try drying grains. I've made a couple of lids but still haven't actually dried a single grain, soon :muppet:
