Awe come on Hobbit, I thought you were the kind tripper to take a ride and write the dosage chart after.

:havesomescience:

My biggest concern is not taking enough the first time :hehehe:

I get that! I test all my cultures to see where they sit then I know it's not going to be one of those rides that you only get a come up and then nothing. I'm never worried about over it's always under.

I would be a lil concerned about that first picture. Yes the for lack of a better term "overlay" and pillowy mycelium is normal for orcha.

But it also appears like you could possibly have a case of wet bubble going on throughout all of that. And that is cause for concern.

I'm surprised nobody has pointed that out.

The second picture looks good though

Do these jars of RW look normal, I feel like the mycelium isn't as white as it should be, lighter grey color almost. I will add i accidently poured an entire 400 ml jar of lc into the 10 jars i got carried away and before i knew it the jar was gone and then I realized how much I actually ended up pouring in each jar, like 40cc lol, They def might be fucked.

I dont like the look of those.

that's likely mold.

Did you test your lc on agar before you used it to inoculate?

Well damn. Thats unfortunate.

You have pics of the clean plate?

The fact that it's already spread over so much of the Grain and it's all wispy and patchy like that..  not really connected in any of the places it looks to trying to take off.  Is a pretty telling sign.

Srry for your loss.

If you still have more of that plate I would start over. Pay close attention to your sterile technique when you  pour.  Best of luck moving forward.:hatsoff:

I think Im done with LC LI's for bit, i have 10 jars of rustywhyte 5 PE all contam pe done with agar, 6 b+ LI jars completely stalled, Im gonnna dump em all outside tomorow, common denominator is fucking oats, Fuck Oats. The 6 jars of rye I bought pre sterilized I knocked up with agar, are the only clean jars I have, and I have 10 Jars knocked with b+ Lc yesterday which Im debating even wasting my time letting grow out or just dumping them fucking too. Going to feed store tomorrow to buy new feed I want rye or wheat.

Nobody will judge you for staying away from lc.

It's super easy to make but still technically an advanced cult skill..

Lots actually recommend stearing clear of it initially.

I'm one of those people. But that just my preference.

I mean there's great uses for it(bags)  and if you want to or need it specifically for something go for it.  Just be aware.

we can achieve good clean results and plenty of fruits without it and while  not working much  about those contanm factors that might come along with an LC.

Especially if your new to the hobby. (Not saying you are)

Hi guys! it's been a while, i lost my account.

I had to close the grow house but i kept some clean (i think) agar and some LCs, now i found a new place so im right back at it, two big issues:
My beloved agar plates (no pour in plastic cups) may have taken a little heat while i was moving everything, nothing crazy, but from a healthy rhyzo myc after the first transfer i ended up with shit

i had around ten of these

first transfers with a sterile scalpel in a SAB

The lc and sslc i made with the same myc of the first pics, tested on agar also looked like shit

after a couple transfers i realized that the room i was doing transfers in has moldy air, since i never had trich in agar plates in the other place and suddently i had 30% nasty contaminations, so i said fuck it and bought a pretty big flow hood (im happy as a child at christhmas), 500 petris and some lc sirynges from a new vendor (last one i bought from had non sterile lcs ffs)

How do you think that could have happened? i was getting healthier myc as i kept making transfers and now this, could it be the heat or the air? plates were stored for around 1 - 1.5 months top mostly in the fridge

thank you

How long ago were those older plates made? Where they dormant for a while?

The change in agar, and from going into hibernation at fully colonized agar plate to expanding to new medium, with different profile, could be enough to change the growth patterns.

Those look symmetrical and with good coverage, it just went from rhizo to tomentose growth. I think you are fine, do another transfer.

Thank you for your reply, that seemed super bad to me, i still have to learn how to tell if the myc is bacterial on agar

The older plates were around one month old, and i kept them in the fridge after taking the pics, it's a fantastic new you're telling me.

do you keep fully colonized plates /LCs in the fridge or are they good sitting at room temp? i mean for short term

Have a look at THIS. It may help you.

I would say that your LC/LI probably had something hidden in it that didn't show up on the plate or you just didn't notice it.  That or  you all at once introduced something to all of them during the pours.

The type of grain you're using makes no difference as long as you properly sterilized it.

That's funny the ones you got pre sterilized are the ones that are performing well. ( but not surprised since those  got the agar and not the lc(

Those are the ones that I would be worried about from the get-go just simply because I wasn't the one to sterilize them and who knows what may or may not have  happened to them In transit.

When you go to the feed store choose something you'd want to work with or whatever is the cheapest.

Make sure you properly sterilize it and then you're off to the races.

Forget about LC for a little bit. while you get your bearings and get some nice successful grows under your belt.

That'll make you smile for sure..

and then you can start experimenting with lc once you're having more success than not and then it won't matter as much if f you fail

cuz you'll already have a stash put away the last you long enough to never care anymore.

Thats just my honest opinion.  Take it for what its worth.      Hope this helps 🙏

Could always cheat and do the "plate scrape" trick I used to do.

1) Get a syringe full of sterile water.

2) Get a nice clean plate.

3) Squirt a few CC of water onto the plate surface and gently scraped the mycleia into the water with the needle tip.

4) Tilt the plate and suck up the mycelia-laden water into the syringe.

It worked for me in the past when I was making shiitake bags in an SAB, but im a sample size of 1 so other folks' mileage may vary.

Also, if you can spare it, make a control jar. IE: make a spare jar of sterilized grains but dont inoculate it. Just to check the grains themselves aren't playing a part here.

For my egos sake I'm gonna stick with its the oats lol. First 10 jars I did of them I used bods easy af tek I think and they sat on a screen for 4 hours till completely dry before pcing for 2hrs at 15 psi, They got contaminated even with a clean agar plate. Second batch of 20 jars I did hams tek I think Where I rinsed them thoroughly and I even threw out all of the floating oats only used the ones that weren't, skipped a soak and just simmered them for about 30 minutes strained steam dryed for about a hour and then loaded them up and pced for 2 hrs at 15 psi, all of them contammed whether it was agar LI or LC, but the rye berries didn't from another clean plate, so I don't know what I'm doing wrong. Everythings being done in front of a flowhood, central air off with a face shield.

I would definitely run a couple controls like the other dude mentioned.

Make sure that your grain is actually properly getting sterilized.

Especially if you're saying the only ones that haven't contaminated on you. are only ones you didn't sterilize yourself.

If You want to really answer your question. Do up some more oats and some rye.(its NOT the oats)

sterilize them. run a couple controls that you don't inoculate and if something grows on those you'll know for sure that's something's wrong with your actual sterilization process.

If those controls do grow mold then you can get some sterilization indicator strips and toss one of them inside one of your jars when you do a run and it'll tell you whether or not inside of the jar was properly sterilized..

Your ego will get you no where in this hobby.  Thats Still funny tho

Other factors to consider are filter failures

And hidden contams.  In the plates appearing to be clean.

That can be frustrating.

You just got to keep track of your process and keep back tracking until you find the actual cause.

Trust me I've been in your situation before. Everthing appears to be clean af but still everything contams.  Same goes

When youve confirmed your grain is being properly sterilized.  then you can start considering other factors and being frustrated with hidden contams.

Yes all good points, I feel like the sterilization process is the easiest part, I vent for 10 minutes bring it to 15 psi and hold it there for 2 hours, The filters i'm using are those adhesive sfds that go over the gas exchange whole i drill in the lid, I never thought about that but I think its not that either, Ill definitely go ahead and make a couple jars up of the oats again and just use them as a test and see if anything happens to them.

Re: Ask Quick Questions, Get Quick Answers [Re: Mortifiied]
    #29642093 -

You definitely got the process down. And it is easy. So if you can quickly confirm that you're sterilization process is on point.  The filters should be good.  I use real sfds with rtv. But the stickers work.

then I would move on to your cultures  and make a couple more plates. Inoculate  do a couple transfers to ensure cleanliness.

Do a couple plates so that you can better odds of success and see if you can get a few clean inoculations.

Or if you end up with contams still from multiple plates that appear be clean.

Do a control as well.

I'm willing to bet its some hidden contam  and  and you just need to  get away from it on the and you should be able to.

Try that.  Come back with results.  Let us know how it goes

Yea i just made 40 new plates and wrapped them I think I might also make a few brand new t0 plates of all the varietys I have too and start from scratch. Luckily I have 2 18 qt monos that are about to start pinning so failure sucks but I'll have some nice fruits soon I hope.
