I just made some PE LC, and I'm putting the natalensis away for now. It's funny how much of a hurry we are at the starting point and then later think "holy shit man, this has kinda turned into a lot and I want a break."

"Hmm, why not put this and this on agar as well, and maybe this while I wait for the first ones?"

Just to find you ran through more plates in a week month than you would ever believe you could need in a year...

I deny these allegations and have no further comment.

Have all of you finished cooking?
I have been invited somewhere and I wanted to bring one dish. Mine is done. It’s just gonna get reheated at the location.

Hope all of you are having a good Thanksgiving.

For actually keeping track of them, the world is your oyster (mushroom). The freezer boxes for those cryovials have nice Battleship-style row/column labels (A-J, 1-10), so you can write down the "coordinates" for each culture so you don't have to take each out individually to find something. I've also just written on the box lid (Sharpie is erasable with alcohol). You can use a spreadsheet, a text file, or a plain old paper notebook. There's probably off the shelf software that would also help with organizing.

It's not Thanksgiving here. Just drinking beer and watching Trailer Park Boys as per usual. Have a great evening though.

Can someone please help me understand whats happening here, I've been into agar for a little over a year. Ive been making my own PDA agar and using ketchup cups with great results. Recently I decided to switch to petri dishes and I tried a MEA recipe:
500ml water
10g agar
8g malt extract
5g charcoal

Now my plates are growing funky and I cant tell if its myc or contam, maybe I messed up on the recipe? Im getting growth but its quite slower than it was on the pda. Some of the rhizo growth just stops and looks like its contamination or maybe tomentose myc? I cant figure out whats going on

Did that wispy stuff grow really fast or did it seem like the myc just changed as it continued to grow? Every time I've gotten mold, it goes from "huh...that's kinda weird" to "oh yeah, that's fucked" in maybe 48hrs. If that thin wispy stuff popped up real quick like, it's contamination. Thats all the advice I have for you - I've never had a plate look like that, but ridiculously fast growth is a dead giveaway to work with.

I've gotten weird growth like that with charcoal. I don't know why - others seem to love charcoal in their agar mix. Maybe some brands/formulations are better, or it works better with a different nutrient? Try without it (or maybe half of it) and see how that goes?

IME, the charcoal retards growth and causes other oddness. I wouldn't use it.

I was wondering if it was the charcoal. I will make another batch without it and see if that makes a difference. I will update back here when I do!

Im still dealing with trich and the last thing I haven't changed is my coir brand. I understand it's the least likely cause, but my jars and agar this round looked OK to my eyes. Pics of the jars are in my journal.

Has anyone used this brand of coir before?

I'll have some proper pics of my agar in about two months. I just found a source of holy grail cups, but they're going to take 3 ish weeks to arrive. I'll post those for scrutiny ASAP. I'm mostly using glad twist and locks, so getting a proper picture is difficult.

Is spore swab to brf puck an actual legit thing to do? I would think agar is your best chance at actually transferring away from contamination

Pucks are good at immobilizing bacteria because of their structure.  Myc moves much quicker than bacteria through brf cake sub in most cases.  You can even do something similar with small grain jars.. before inoculating, hit all the grains to one side, shoot the SS on the side without grains in it.. then hit the grains back on top of the SS.  Let the spores germ and don’t shake the jar at all.  The myc grows up towards the top and any bacteria stays at the bottom.  When the myc reaches near or at the top, open the jar and pluck a colonized grain out and transfer to a new jar.

Mold is a different story and everything is probably going to be easier and more reliable on agar but there are ways around not using it for sure.

My plan was to transfer away a plin to agar, but the Nats I did seemed to stall out. I think my mixture was on the dry side.

The main reason I'm doing swab to brf pucks is because the last time I did a swab to agar I ended up with plates that were more bacteria than mycelium. I spent months trying to clean them up, but couldn't get a clean culture even with water agar, josex poke, etc...

FWIW, it seems to be working. Here are some pics from some glass petris that I transferred brf material to:

They will still need a couple transfers to clean up, but as a starting point they seem to be in much better shape.

Edit: I'm thinking of taking the following transfers but any opinions are welcome:

Pic 1: 9-11 o clock
Pic 2: toss
Pic 3: 6-9 o clock
Pic 4: 3-4 7-10 11-12

Edit 2: I just realized Pic 2 is agar to agar not brf to agar. I must've mislabeled it. Still going to toss it.

Besure to just take SMALL pieces  from leading edges...but sounds like a solid plan.

Also for the bacterial situation don't forget about hot-pour/sandwich method, for the arsenal.

Especially with glass jars you've got more room to work it up.
Like first top layer being WA maybe 2 layers to be sure your cut doesn't grab the bottom layer.

*Also maybe a little higher % of agar helps to immobilize bacteria.
0.2-0.6% more agar makes a huge difference.

Definitely. Like faht said, it’s great for bacteria, but I’ll just add I have used them to clean up very moldy swabs too.

Got some swabs that were 11/10 filthy. Couldn’t germ them on agar and when I did it was all green. Jamming the swab, which I hydrated with a drop of sterilized water, into BRF got the cube spores in there to germ. It also got the mold spores to germ, but there was a very clear division between the two and I was able to make easy transfers to agar to clean it up.

So if you’ve got something dirty/stubborn/both it is a great tool.
