You don't need to let the culture come to room temp first. It'll need to warm up before really taking off but it will do that just fine among grains.

Not necessary to wrap jars. Unmodified lids can stay slightly loose for gas exchange (the threads seem to prevent contamination, just make sure to tighten before shaking or agitating). Filtered lids should be tightened.

Rookie researcher, trying both pf and tub tea. Yesterday I took a cpl pics of my most aggressive colonizing bag. It looks just about ready for s2b, but being my first shot, I hoped to get some advice, as I do not want to s2b prematurely. This pic was taken 4 days after the break n shake, and is only 17 days after inoculation. Today it looks even better, but this is the most recent shots I’ve got handy. Advice very much appreciated ❤️

Hi, new grower here, just checking to make sure that everything is going fine with my grow. I thought it was fully colonised so I misted it and moved it to a cooler more well lit environment, but it's been about a week and a half now and it hasn't really changed at all. The images above are pretty much exactly how it looked a week ago.

sorry, one of those photos was meant to be this one, still getting used to the forum.

Just an update, the grow bag is going well! Got a few mushroom pinheads having come up overnight. Only thing is, some of them are growing off of the bag. There’s a bit of a mesh of mycelium clining to it, and some pinheads are growing from there.

I’m thinking I should remove them because they don’t have much stability, they likely won’t grow much, and they’ll take resources away from other mushrooms.

In most cases it comes down to dry grain or shitty cultures

I feel like I've experimented so much and realistically eliminated so many variables at this point that I wonder if it actually could be dry grain. I have generally been doing no-soak in all but one attempt, which involves boiling until a few burst grains and opacity all through the thickest grains. Then I spread out to steam off, load and PC.

My impression was this should be adequate hydration. Just about willing to follow any lead though at this point. People will point to culture and sterile technique, and I get it, but I would happily bet my house at this point it's the grains. I've just tried too many different cultures and emphasized sterility in the SAB too much to have the same exact problem every time in every jar.

This is probably a stupid/obvious  question but I still need to know? When working with agar and Petri dishes inside of a sab,  do we spray and or wipe the dishes with 70% ISO before opening and working with them like we would do everything else?

I'll be real, I don't really do any of that anymore. I use an iso paper towel to wipe debris off my scalpel blade when doing dish work, but otherwise I don't use it at all. And I really don't need to use it there either.

I use a second trivet, but I doubt it's necessary, as long as the cooker is full so nothing falls over.

Lids loose, tighten before removing from PC. You can get away without loosening filtered lids, but cranked down plastic lids are (in my experience) more likely to warp or to suck in water that happens to pool on top, especially if the lids are already warped. Foil mostly prevents that issue too.

I don't bother with foil for unmodified lids, but it doesn't hurt.

I don't think I will ever stop wiping stuff with an alcohol-soaked paper towel or cloth. Maybe it's not necessary, but it's a really easy measure to wipe off any loose mold spores. Water might be just as good for that but it doesn't evaporate so readily.

You don’t need to use bleach or isopropyl alcohol in you’re still airbox. You could just use soap and water and wipe stuff.
I just like isopropyl alcohol, and I have a lot of it.

Is it safe if jars are pretty snug agains the walls of the pc? Im kinda paroinoid about pc'd and the added pressure of the glass pressing on it. Obviously didnt bend it or anything. But its tight. I did mu top layer as a triangle. But one was wodemouth and its kinda funky

Okay so if I'm working with a sleeve of pre poured sterilized agar plates dishes. I should open the sleeve outside of my sab? (So I don't disturb any of the still air inside.)

Place all the dishes inside. Stacked or unstacked? Then one by one remove lid inoculate put lid back on. Then wrap all at once at the end with parafilm? And remove from sab and place in storage?

This is going to be my first time ever working with agar so any advice would help.  Thanks in advance.

I like to recommend Bod's videos to anyone who's just starting out with agar. Even if you don't read his posts that detail his agar workflow, the videos should be enough to give you an idea of how it's done.

To answer your questions:

  • I personally like to spray the sleeve with iso and remove the plates inside the SAB but I genuinely don't know if this is a good idea or not. Works for me though so it can't be too bad.

  • You keep your plates stacked - see Bod's videos.

  • Whether you wrap the whole stack at once or you wrap your plates individually is up to you. Think about how you're gonna use them and decide accordingly. I personally like to wrap them individually.

NO!
If it is sealed, then it is sterile inside of that package. What you can do is spray with isopropyl alcohol place it inside of your still air box. Wait for it to dry after wiping it off then you can unseal it.

It's been less than 24 hours since I inoculated a PF cake, and there's already a really, really small white spot around one of the spots where the spore syringe was emptied. It's hard to tell because it's small, it might be a really light colored piece of the vermiculite-rye flour mix.

Has anyone had visible growth with PF tek this quickly?

Coir needs to be pasteurized for bulk substrate. What about for casings?
