Hey good to see Beef on the grain train   I guess the more I think about it, the more the blood thing tracks.  I've thought that they had notes of minerality (as vague as that is...), which runs a parallel.  They also have notes familiar to cube myc and a foresty/loamy undertone.  Referring to the spawn specifically here.  Been obsessed with the smells of different exotics lately.  Semilanceata and caerulipes are divine.  Semps are complex and ominous.  Caerulescens and mexicana are something that should be bottled and consumed, frankly (I love melon/cucumber notes), samuiensis smells fucking weird and potentially toxic...  A real rainbow of nose ticklers. :mushroom2:.
Yeah I was wondering also about how well the grain capsules would store, and the safety profile for long-term storage.  You're definitely dealing in a different nutritional makeup than with fruitbodies as far as how/what bacteria and fungi might take hold given the accidental introduction of any moisture.  Still rad AF, so I mean . But worth noting...
Going to be aging and testing papuana spawn once I run the BWT-01 line that tested so high for Wavy.  Have that on T1 now so I'll have that going soon.  Thinking maybe caerulorhiza as well given the insane blueing seen on the agar plates.

My bad, I thought you were testing bisporus mycelium, not mush.

Just rereading this and I think I missed your point. Yes, I will do some bisporus grain once I have a culture that I trust. To me that trust comes from a culture that I know fruits. Otherwise, I guess I could be testing mold or some other contaminated mess. Ideally I like working with a 2nd gen clone, but if I have a culture that fruits well, I feel good enough running tests on its grain spawn.

To be fair, some tamp cultures don't fruit well but do produce potent stones in the mycelium, so my fruiting culture theory could be total crap. It just gives me a sense of confidence, misplaced or not.

The new bisporus cultures from bra are looking really good so far on agar, so I am hopeful.

and we have a product.  interesting.  2 qts of grain came out to 312g or over 11oz.  yet, the capsules weigh approx. twice as much as a normal capsule at .49 per consistently on a #1 cap.

my theory right now is that theyll hit about the same as a normal capsule but weigh twice as much.  (but you get twice as much per quart)  Im not sure what to do with that info just yet .

Testing begins this weekend.  it sure turned a deep blue in the nesco so im thinking its gonna slap pretty good.

Down to my last two jars. One went into the dehydrator this morning.

They were beginning to look kind of ~~~ with a gray layer on top. I suspected that they are getting too dry because the jars are pretty light, but they still felt a bit hydrated when spreading them on the tray.

The one on the left is lions mane of approximately the same age for no particular reason.

Bit of bluing in the couple of minutes from shake to spreading on the trays.

Plan to update tomorrow with results for day 306.

Okay, I am lost.
This sample looks darker than the last. And the Colorimeter bares that out. The problem is now that I know I was misreading the scale before, I can't make an adjusted estimate of what I should call this.

Using the colorimeter on two independent samples last time I discovered that I was vastly overestimating the results. However, they were consistent with each other and with prior tests. So while the quantities were wrong, the curve was valid. Now after what appeared to be a clear leveling pattern, I am confronted with a significant increase in potency confirmed by the absorption meter. So I guess the only thing to do is apply the percent increase from the meter to the incorrect data on the graph to continue. And more importantly, I should start the whole experiment over with new grain and always testing the samples using the colorimeter.

Enough whining. Here are the pictures and updates.

Dried grain

Dropped another 7 grams to 45 so the myc is still alive and eating.

So it is noticeably darker than what I called 12mg/g before getting the light absorption measuring tool. But I now know that what I called 12 was actually 1.29. So doing some sloppy math, (1.79 - 1.29)/1.29 = ~38.8% increase in potency. That result multiplied by the incorrect estimate used in the graph gives me a new relative potency of 16.65.

Of course that is not correct, just an extrapolation of my initial errors. After I get the final measurement in two more months, I will use the results to reverse the ratios to get a more accurate scale on the graph. But the curve should not be affected by the change in scale.

But now lets look at that curve. Why in blazes did the potency go up? The total alkaloids is remaining constant as the mass is reduce while the percent of actives increases. Cool, cool. But the jump is unexpected.

So in conclusion, this whole thing is trash and I will redesign the project using what I have learned, better tools from the start, and now it looks like we may need a longer time line.

More homework! Who said that drugs are fun? On the plus side, Miraculix now offers cheaper test kits if you buy in bulk targeting the colorimeter.

Fuck it man so long as the potency variation is correct sample to sample, that’s the most important part.

So theoretically you could start again and take an accurate reading at the first stage and then should be able to predict potency at each future point. Pretty badass.

The latest jump does seem odd, maybe for some reason after so long it began to break down the grain and reduce the total weight somehow. Or you waited long enough to unlock the next level

Assuming things go as planned I should have some extra money by the end of summer, I’ll kick in on a batch of tests for you as soon as I can if you want.

This is such an interesting area, it kinda seems like most other stuff has been 99% figured at lately and this is the only unique thing happening.

Necromancy but possibly worth it. Haha

Has anyone considered using sorghum as the grain/substrate for this method? Within 6 months or so the mycelium will have consumed nearly all of the substrate and, theoretically, one will be left with only active mycelium to consume/test.

my last batch, 90 days wasnt quite what i was hoping for but are still very useful.  i made some #1 capsules with ithe flour and theyre kinda nice.  1 is unnoticeable but 2 is def there.  so you can really dial in a semp dose which is difficult to do otherwise.

i have another set of jars i think im going to let roll for 120 days which will be this time next month.

I'm not sure if I missed it but were these tests done with the fast or slow var? Someone in the subtrop thread recently said that slow var is more potent and that fast var is the potency of some cube. I would think that subtrops are more potent in general than any cube, but I haven't done enough bioassays to know for sure.

Why would sorghum be better?
Wheat tastes nicer for one.

Well, there would be very little "grain" left so it would be easier to calculate dose and consume; much like dried fruit bodies. Also, seems like the "discoloration variable" possibly attributed to grain in the tea/test water would be diminished or eliminated entirely. Granted, it would be likely be the final alkaloid peak since one would have to wait 6 months or so till all the sorghum would be consumed.

I believe it's what Aloha medicinals and other companies use when they're after medicinal mycelium. Ground up and put into capsules being nearly 100% mycelium without substrate 'fillers',tedious separation tasks, or gluten allergy concerns. -shrug-

Nice Yoshi! Good timing as well! 👍🏻 That was going to be my next question, for real; if anyone had any experience using Milo with sclerotia producers and their experience with it. Mexicana is my personal favorite. I have yet to try subtropicalis but they're on grain 🌾 now and the countdown has begun. 😁

I'm not sure if anyone has attempted to see if mexicana (I've read of one account in which tampanensis works via this method but unsure of truthfulness) also has alkaloids in the mycelium but I can bioassay next week and chime back in if anyone is interested. Or tonight if the spirit moves me. ☺️🐌🤣 They don't stain blue like other species; probably due to lack of psilocin so the only way to know is to dive in I suppose. Haha

Pan Bisporus runs on hull less popcorn as well as tampenesis does on grains.

So popcorn is a viable substrate for myceliated pan grains.

I'll likely fruit what I have now, and start some new jars to try out some myceliated grain spawn.

I'd still like to know where this very little grain left statement comes from.
Have you grown with sorghum before?
What makes you think the mycelium will eat all of the sorghum because it's just a grain like other grains.

My tests for the graph so far have all been from a clone of the "fast" line. I now have two of the slow lines nearly isolated enough to do some side by sides comparisons. So that will be coming down the pike. So far, there is even a large difference in the speed of the two "slow" genetics, but I have not had a chance to test either by QTest or bio assay yet to add to the which is more potent discussion. Once I have a reliable culture of each locked in, I will do some side by side testing keeping all variables as close to identical as possible (grain, prep, temps, chambers, etc.) and test myceliated grains' and fruits' potency for those lineages. If I start a new post for that I will link it here.

I am also curious to understand what it is about sorghum that leads you to believe that it will be more digested in a shorter time than another grain type? Not saying that it is untrue. I would genuinely like to know if there is a better performing grain.

LC has been proposed as a possible method of growing pure mycelium and I can see that as a method of making all of the nutrition more available to the fungus to consume completely. Finding the most efficient nutrition density formula to finish with maximum mycelium to residual nutes would be an interesting line of study. Kind of like the hydroponic equivalent for plants, but for fungus.

Yeah man lc would be the way for pure myc. Some of the mats I’ve had form were huge, and I’m sure if you poured it through cheesecloth or bubble hash bags you’d catch most of the rest.

A while back I was considering making natalensis lc in half gallon jars and then laying them on their sides so a huge mat would form but never tried it.
