Grain that doesn't take...

I had a grain bag that didn't take with some LC. The other 2 did. Could I put the bag in a PC for a bit to resterilize and re try inoculating?

Didn’t take? Like the LC didn’t end up colonizing any grain? Did you use enough of the liquid solution? Pictures can help a lot.

Honestly, I would just start over. Maybe reuse the bag? If at all? But if you have a PC, why not just do some jars? Easily reusable and overall great for the environment.

I think I put around 3.5cc into it. Its been a few months. There is nothing to show its just a normal grain bag that looks fresh. No contam, no moisture it looks brand new. The mycelium just didn't take I guess.

I have switched to using grain jars and buying grain in bulk, but I have a few grain bags laying around still. I have a bunch of LC so I think I'm going to do an experiment and PC the bag for a bit and re inoculate it. Worst case ill just have to throw it out later.

Looks like you have bacteria riding along. I would consider taking from the leading edge? And transfer on to a new plate. But could also consider getting a print from the marketplace? Prints are easier to work with IMO.

Just swab and swipe.

If you insist on using the syringe, make sure it’s thoroughly shaken and only apply 1-2 drops. That should ensure a good chance.

Do you have other germination plates? How do you make your agar and with what nutrient? What are you trying to germinate ?

It states that it’s LB agar. I foolishly bought them on Amazon. I’ve since bought the materials to make my own. I think I’ll just trash the plate fo safety sake. No action on the other plates I did so maybe no spores came out. Always learning.

I got some of these: "AirPort" Lids for LC and Grain Spawn

They ain't great for grain spawn

Yeah, the hole is really small. I'll go back to flipping the inner part of a normal lid and leaving it slightly loose.

for those who have experience with long-term storage of plins in cryovials with sterile water: it's normal for them to turn completly black? - thought this kind of oxidation would need oxigen... starting to doubt if the cryovials are air sealed...
