about as well as can be expected so far.  didnt get any germ on the maak+F2 and yeti dishes.  just put those 2 to fresh agar again in hopes i get germ this time. used dishes that still had some excess moisture in them which i hope helps this attempt. should.


i recently did a hard reset on what i'm running genes wise.  got a lot more plates spinning than normal. been doing a lot of the "secure clean cultures shimmy" lately with a bunch of varieties. i'm excited to start playing with some of the newer genes making the rounds recently. my library is mostly older school stuff.  thanks again.

its ape so a bit slower is expected, but this is also a really old generation 4 clone.

How long can you really run clones of clones before you see senesence ?

I disagree. When you make a transfer or a clone, think about what you are doing. Germination greated a cell line the propagated by cell division. You transferred from that cell line which also populated only from cell division. Every cell division adds to the likelihood of a genetic mutation which will cause the cell line to destabilize, aka senescence.

The only thing that resets this is fruiting and germinating from fresh genetics.

The reasons people tend to say we wont see senescence is because we store a single master in cold, and take transfers from the same source for a year or more before having to clone the master. This means every grow out comes from an early generation, thus it never gets old via cell division.

If you keep taking transfers of a transfer, your cell line will eventually die off. That eventually is likely in the hundreds, thousands, or higher transfers, but a very unlucky transfer from a cell line that had a random, but deadly cell mutation will die off much quicker.

The odds that you will see it by transfer 4 is miniscule. You should be able to go for a long time before having your cell line die off.

Ok you got this all wrong man.

A clone contains multiple genetics (unless it was from a true isolate which is pretty rare).

No senescence.

You are conflating having multiple lineages in a transfer vs every single of those multiple lineages only has one way to propagate, via cell division, which started at germination.

Now, you you know that these cell lineages are diversifying while growing out and intermixing there genetics, then I'm definitely wrong and each time that happens they are 'fresh'; however, I'm fairly certain that only the spores create fresh genetics.

So at which point do you guys take your first sample for storage? Ms -> Clone tissue -> clean out on agar -> longerterm storage? Or before longterm storage spawn that clone tissue and take from the flush an nother clone before putting that into storage?

I've yet to find a clone worthy of long term storage, I'm still going from MS to clone to MS. However, I would only consider long term storage of a clone that I knew grew out amazing.

I grow the spores and fruit a tub. Then take a nice clone and immediately slant it from the very next plate so long as it is clean

Otherwise I feel like you are just capturing the MS in your slant. So you could just start again from spores you made from the grow, why slant it at all. I feel like taking a clone narrows all the potential outcomes. However I can't say the clone always represents the parent that faithfuly in my experience :shrug: but how else can you do it really

This is my second attempt at making agar. First attempt at spore print to agar. I believe I have some decent mycelium growth, but am a bit concerned about the thing clearish layer surrounding it in some areas. I think I should let them go for a few more days at least and then make some transfers.







Opinions appreciated.

Sooooooooooooo tonight I used a Corning media bottle for the first time instead of my cheap hope made bottle with a GE hole.

PC'ed the Corning media bottle with the lid a quarter turn loose. When I got it out of the PC, I tightened the lid to shake the liquid inside... and left it like that.

Comes the time to pour, I start unscrewing the lid and hear "pfffffffffff". Sucked a shit ton of air in. I'm expecting those plates to be goners. So I was supposed to let it cool with the lid not fully tightened I assume? :shrug:

If you did that in front of a flow hood, it should be fine. Maybe in a SAB too. Where did you do it?

If not, might even be OK still, sometimes just out of the PC they're still hot enough to kill nasties.

If you aren't pouring a lot or don't mind wasting plates (I order them 500 at a time for 110 dollars a box), I'd say pour them anyway, and then  tape them up in a stack of 10, put back in the sleeve, tape them up, and leave them at room temp ten days before using. I do that anyway and I pour in front of a flowhood.

I PC those Corning media bottles with lids very loose. When done, I let the PC sit on the stove and pressure drop to zero, then bring the PC with the lid still on into the pour room, take out one at a time, and close the top to keep nasties out and also keep the agar from solidifying.

I've never ever heard air sucking when using one.
