well, I am just handling it like I learnd it when I was working in the lab, won't see anybody there boiling agar :p But yeah, many way lead to rome ;-) I just tend to stick with what I'm comfortable with.

I still heat mine in the Michael cave before PCing. I know it isn’t necessary but I had such tribulations with agar that I’m just going to do what worked until I’m no longer skittish lol

Ok so another quick question if you guys don’t mind…if I have a plate that is clean and fully grown out but I don’t have a need for it quite yet…can I store it in the fridge until I do have a need for it? Or what’s the best method of storing a plate that’s colonized and ready to go to grain but you aren’t ready yet with the grains…and with said storage method…what is the life expectancy of it? Like how long can I store it? I’ve read all kinds of different answers

An agar plate can last months in cold storage, and that is the preferred method, yes.

Just do a transfer of it to not worry about "time" and room temp is fine for agar plates...

2-3 months before it gets dry it will last...

I don’t have an insulated lunch box…right now at least…do u think I could get away with parafilm on the plate and then putting it into a ziploc? To prevent condensation…and when they are just in storage with nothing in them I store them upside down…do I do that with colonized plates as well? Thank you guys for the answers!!

Do you guys think adding a fair enough amount of activated charcoal to agar or even lc would make it podible for the culture to be stored for longer periods?  I heartd pasty saying adding wood does that

So I had made a post a while back praising the GWA I was using. My spawn started off looking as clean as anything I have ever had. I thought I was doing great...

That is until I started shaking the jars of grain, and it's now evident that ALL of my spawn is bacterial.

I felt pretty good about my SAB technique, and I can't see contaminating EVERY SINGLE jar I have. (No molds that I can see.) I think the GWA wasn't allowing the bacteria to thrive, making it invisible on agar, but present.

I'm about to pitch over 2 dozen mycoquarts of grain! Everything I have on grain right now looks bacterial. My plan is to switch back to a traditional agar recipe, and start over from spore on everything. :facepalm: I guess I should be happy I didn't waste a bunch of bulk substrate as well...

That was not cause by GW but subsequent implications.

Let me explain how I see it.

First of, you need to PC GW a little longer than LME (30 min IME is fine). P9 I think explains the whys in a post I can't find right now.

Second, I recommend making GW from boiled grain water that hasn't soaked prior. Soaked grain will start to ferment and release a ton of nasties you don't want in there.

If this isn't how you were doing it, change those 2 things and love GWA again :super:

Sounds like the same problem hazyhorse, I and others have been having. Initial tests are looking good so far the problem is how you're running the PC with grains. Especially if you use a larger PC. You really have to step up your vent time. All internal and external temperatures of the load and system need to be the same and venting before adding the jigger on to build pressure. P9 talked about it in his verification thread, he vents his x75 PC for 30mins, I started venting my AA941 for 45mins. When I did my bacterial issues disappeared.

So if you have some thin walled presto that heats up and starts venting in 15 mins there's no way the load inside is also heated up equally and vented in the next 15mins after that. Then you add the jiggler and trap those colder pockets inside the middle of jars and they don't sterilize completely...super close...bit not completely.

My grains don't soak, and soaked grains are sprouting, not fermenting. I think that's a misnomer that is set in stone on the Shroomery, but I'm not giving up yet.

PCing for 30 minutes instead of 20 is news to me, but I can't see how that would change anything. I'm doing pre-pour in jars, so there's no way 20 minutes at pressure isn't sterilizing everything.

In control plates that were intentionally contaminated, I saw some bacterial colonies, but they always stayed small. Plainly visible, but small. IMO, bacteria should spread quickly on agar. Stored plates never showed any signs of contamination.

For now, I have to switch up my agar nutrients, because I can't even test my SAB sterile technique unless I have agar that will readily contaminate.

Hey :smile:  because my life can be pretty unpredictable so I can never be sure if I'll just leave some transfers I want to do now pending for a few weeks or even months, although I had some caltures produce fruits in agar water after 2 months of not checking on them xD
