That's a good idea foam block with holes..

That should stabilize the temperature swing of a refrigerator.

I don't know how many people are using 50ml. centrifuge tubes for slants.

I use those as well, though I have noticed some varieties simply will not grow in a slant if it has the wood popsicle stick inside. It's the strangest thing. Some love it, others won't grow at all. You knock them up and the piece you put in there won't even fuzz up, let alone spread to the agar in the slant.

I haven't heard or experience that. Thanks for the info...

I got that bag of 50 too and ran through them all. I tried to no pour them. Turns out the blue caps are not polypropylene... The green caps are. I just stack mine in a soft sided cooler with an icepack on top and bottom.
I'm starting to use Mr aliens holy grails poured super thick with some sawdust pellets broken up in the bottom as psudeo master slants

I dunno why that would be the case but I use little bamboo skewers cut to size (the ones with a sorta rectangular profile to the skewer for making fancy satay sticks), and have no issues of that sort

love those 50ml centrifuge tubes, but I always used them and have no experience of anything else to compare

Thanks for the bamboo idea Nef. I’ve wondered if the reason for what I’m seeing has something to do with the type of wood my popsicle sticks are made of or potentially what they might be treated with. The ones I use are birch. I’ve been wanting to try another type of wood to see if there’s a difference. I thought about coffee stir sticks but I like the bamboo idea. Will give it a shot!

New bootleg bacti-cinerator.

For those that are working with these, do you need to keep the tube clean? I’m thinking black carbon build up from scalpel blades will collect in there. Do you do anything about that?

I just use toothpicks.  What species have a hard time with colonizing your slants?

The ones I bought are birch. I'd say 50% of my cultures will grow on it and 50% die immediately. Strangest thing. I like the toothpicks idea too - easy to find those!

Yeah, don't use dirty tools with those. Keep your tools nice and clean, try not to scrape the walls too much, etc. Carbon doesn't build up on the blade in the same way that torches do though.

If I notice any excess agar, etc, on my scalpel I give it a good wipe down with iso and paper towel.

Watch your fingers, if you leave the scalpel to rest inside of it to heat while working you'll wish that you hadn't.

I know the ones you mean, for levelling cement and the like? Yeah pretty steep for what they are. That’s why I opted for this as I almost exclusively run shoeboxes at the minute and this is the perfect size! Set me back about $4 from Bezo

That’ll work perfect.

I always thought about using a trowel but it just wasn’t that big of a deal. Then I had to buy some for a project and tried it out on a few tubs. Works way better than I expected, especially on big tubs.

Good to know! Gunna give it a whirl later. Got 1 tub to spawn using some left over CV from the other week. Looks and smells okay and has been locked in the ice chest for the duration. What could possibly go wrong?

Also drilled out a 22 litre samla mini-mono yesterday ez dialed style. Curious to see how that performs….
