When using a sketchy innoculant in this case lc it is best not to mix or spread it around. Shoot it in a concentrated spot. If mold grows in that spot well your lc is crap. If it grows in another spot your grain or sterile technique is to blame.

If it's all good you just break up that clump and spread it around.

thanks for the advice, I just squirted a bit of that same LC into 3 different agar plates today using a SAB. I guess we can find out in a few days. Ill wait till then to shoot up the rest of these bags. About how long will it take to see anything in agar by the way? and can If there's mold and mycelium growing, can I cut out just the mycelium with a scalpel and transfer it to a fresh plate? Is this how you clean it up?

Yup that's how you clean it up, just transfer away from the bad stuff.


It'll probably be 3 or 4 days before you get some nice growth in your plates.


You just did one drop per plate right?

I don't test my lc. on agar.

I find that It's easier to spot contaminates In lc's than a dish. If mold is not in It's vegetation state It can be very difficult to see..

An agar plate can look clean and be dirty with hidden mold. and you pass it on to your spawn jar, and your colonize grain will look clean. Then It vegetates in your substrate and you loose a tub..

In my eight years of working with lc's I rely on my eyesight and my nose.

If you have turbid lc/broth, you have bacteria (not to be confuse with sediment that you stir up from your formula.)
It should be clear when you hold It to a light source.

If you have mold It'll float to the surface and you'll see a white spot floating around then it will vegetate out with time.

Normally sediment will diminish with in a week as your culture consumes the food.

If your liquid culture floats to the surface It could be starved for oxygen, so It's a good Idea to give It a short stir to break the surface tension and let the lc oxygenate.

Can you spot the test tube that has bacteria?

Oof yea buddy you made this wrong. Agar first then LC like others have already said. You can test it on agar but I'd bet its contamed. Noone can tell you just by looking at it if its clean, only testing will do

I always go agar first, then to my broth..

I make small 130 ml. batches of broth per jar to receive the culture.

I use to test my lc. on agar but not anymore.

You're always taking a chance of contaminating your lc just to test It. Then how many days are you suppose to wait?

Mold doesn't always show up on your agar plate.

I had mold show after 6 weeks on a set of plates, they all showed green on the same day. Mold has away of hiding.

I would purposely grow mold in my lc. broth just to study It's behavior and see how easy I can spot It's growth.

From what I have observed Is that mold will float to the surface as a small white speck and to a sizable glob at a rapid rate, then vegetate out.

I don't know that much about bacteria but If my jar turn cloudy turbid looking I toss that jar. If It's clear I go for It..

I personalty think too many people read to much into it.

If It's turbid or white blobs floating on the surface toss It.. Other wise go for It and learn from your mistakes.

The life span of my lc's are probably 15 days. I normally use It on the 11th day or so..

Hey micelio

What about very slow growth? I have batch of 5 LC's that I made on the same day and 4 are taking off and looking good. One is very slow though — no cloudiness or white spots, it's just very slow to expand.

I'm wondering if this can be a sign of contamination too?

It probably not contaminated. It could be just the amount of culture you added to the one jar than the others..

I have had spawn jars do good and some grow slow from the same 60ml. syringe with the same lc..

What about these? They’re 20 days old. I’m concerned because they are not so clear anymore. I put some on agar 2 days ago so we will see.

Do you see the mold on your upper portion of your jars?


Mold growing in sterile water.. They all turned green

I see the cracks also, top and bottom center. Is that mold growing on the glass? With LC it gets pretty hard to see if they are clean, but they don’t look right to me. Granular, and some growths on the glass.

I can assure you there are no cracks! Haha although it may appear so at first, the jars are actually just heat sealed with cellophane plastic.
