seriously. i can't for the life of me get the proper moisture content in rye--i've tried five times in the past 2 months and haven't succeeded once. yet with extremely little care or attention to detail, i can get quart jars of distilled water and honey to colonize in just a few days! i didn't even measure how much honey i poured in the jar, nor how much water, and i simply inoculated them in the open air, with no mask or gloves, only a minimal amount of lysol spraying. i did use alcohol on the needle...hmmm. do you think the fact that my liquid jars are only covered with a few layers of tyvek held down by a couple rubber bands has anything to do with my success? that is, do you think that my rye jars weren't getting any gas exchange while in the incubator and that my liquid jars are getting optimal gas exchange, hence their rapid and successful growth? i normally left the rye jars covered with foil and the lids screwed on tightly. i had been told that gas exchange really isn't a problem in the initial phases of grain colonization--it's only beneficial in the latter stages of growth. hell, maybe i should try rye one more time. it was so damn dry that with the added moisture from the mycelium culture it may just work. what do you guys think?
also, how long is too long to keep a liquid culture in the incubator? i've had a B+ culture in there for 3 weeks now. i'm not ready to take it out and shoot up some jars. i think i need to make a glove box for the liquid extraction and the inoculation of the jars; i'd rather, not but since i'm 0-5 with rye (even though contams haven't been a problem and i'm not using rye next, it'll be wbs) i don't want to take any chances. should i take the B+ culture out and put it in the fridge until i'm ready to extract the mycelium? thanks dudes.

I dont think your problem is with moisture content. You absolutely need a polyfill or tyvek filter,I like polyfill allot better.

i do. i always have two layers of tyvek beneath the lid. but you could be right, i dunno. i never noticed a big contam problem, only when i used anno's no soak & simmer method of just measuring out the right amount of water and grain and then pc'ing did i have contams. of course, on that occassion the jars also showed what looked like the most optimal moisture content. then they contaminated.

Its not difficult at all
soak 12 hours rinse and simmer 30 mins let it drain for 45 mins load in jars filters and foil pc @ 15 psi 90 mins
And if you do that exactly like that, and inoculate in a glove box sprayed with Lysol you will never get a contam. I get like 1 in 100 jars contam on me, and thats probably because of a dirty print.

Use brown rice, mahattma store brand. Organic grains contam when not soaked.
No need to soak, mix the water rice, and then pc.
Ratios are VERY forgiving bacteria/contam/mycelia growth wise. I could post pics of AFOAF's rice right now of bone dry and sopping wet doing fine.

I would try this if your having problems just loading and Pc'ing the grains. Soak -4- life! :P

not to be rude, but you simply dont do the same volume. that is simply too much work for huge volumes.

hey guys, while i appreciate all the help with rye, that's not really what i'm looking for in this post. anyone out there have any comments/answers concerning my liquid cultures? *see first post*
