Talking to some agar friends, if you really want to expose some meshed in mold use YPD yeast peptone dextrose agar. Then clean it up on water agar. I haven’t tried it yet, next paycheck I’m buying some.

That's cool, how does it work? Do they grow differently or separate more clearly on YPD?
Heard of V9 agar recently which is just agar with V8 juice and some dehydrated potato flakes. The low pH inhibits bacteria and it's also supposedly a good medium for germinating stubborn spores. Might try it on my next germ plates.

It’s a really high nutrient medium that things just explode on. Allegedly makes it easier to identify the separate organisms on the plate and isolate out the myc. Then when you transfer to 0 nutrient water agar the myc has a chance to outrun any small amount that may make it with your transfer.
I’m speaking from side conversations though. I haven’t tried it myself and certainly not an authority. It’s pretty expensive so makes sense that it’s not in everyones SOP’s

I've got a PESA that does that heavy as well as an Azzie and an Ovoid. Could be Tam's. After many transfers and new generations from spore I think that is a less than common but perfectly normal phenotypic expression and not always indicative of other competitive organisms. Pheno expression is often influenced by environmental factors such as competitive organisms and nutrition etc. That pattern seems like a response for sure. Shit definitely acts weird when other critters are around. If anything I would suspect a mycovirus. Mushroom AIDS.
It freaked me out the first time I saw it and I spent hours and hours staring at it and watching it under the scope. The mycelium in the thin patches seems mostly subterranean and barely comes to the surface but looks identical to the other mycelium and clamps and joins and separates with it like it should. That Burma 4 generations later (spores) does the same thing pretty regularly.
I like and look for weird shit though. Rarely do I run normal looking mycelim anymore as it leads to normal looking fruits.
I'll swipe some burma soon. I got bored of it after a while but I guess it's worth a revisit.

Well I've already got other tubs running and simply for fear of contaminating my entire room with more spores and ruining other projects, I tossed all those plates and grain jars containing that gen line (plus 2 tubs that I'd already spawned, but not fruited so hopefully they didn't contam my house if they did indeed contain mold.
I must admit I was really tempted to just use all the grain and see what happened, but my paranoia took over and I tossed everything. Would have been interesting to see.
My last tubs all had vert (same gen line), so I'm kind of assuming it's vert but not sure. Didn't show up until 3-5xfers tho, whatever it is.

some side-by-sides of 5G LME to 10G AGAR (blue) and 7G LME + 1G YEAST to 10G AGAR (green). Testing + and - nutrition to see wtf is going on with my plates. Also cookin up some BRF now to compare side by side to LME. Both for ~410-420mL water
some mak T1s that i haven't decided to transfer from or put to grain yet, i think a few look ok. What do you guys think? I'm looking to fruit as soon as possible to get a clone to work with

Those are looking pretty damn good to me moshe. I'd probably go straight to grain and take another transfer at the same time in case the trich hits the fan.

You can transfer most of them now/soon besides the BE which hasn't germed yet? Hard to tell from the pic. You could give the slower TAT and MAK plates another day or two but if you can grab something now no real need to wait. Worst case if you don't get good growth you can just go back to the germ plate n try again.

BE is actually PE (long, sad story) and two have germinated but it's not readily visible. Ok, I think I will them another two days if they're just about at the minimum for transfer. I think I'm going to make a shared louisiana albino + TAT plate as well.
Anyone ever use yeast for a liquid culture medium? I was planning on making some BRF LC and was wondering, since yeast seems to be successful for mycelial growth on agar, if this too might be a good idea.

This is the strangest plate that I've got, Cambodians. It's literally the 4th "flush" from this petri, I pick them off and clone them everytime and it just keeps going. Strangely enough I've only ever had one transfer show contamination.

I get those often when I'm isolating for vigor on early transfers - so it's been common in my experience.

@DERRAYLD That plate looks crazy, first pic almost looks like an old tampanensis plate. Maybe it's just the lighting, but very cool that you've gotten multiple flushes from a plate
@moshe1 Same experience as Mel stated. I get this very often on my T1 or T2 plates. Then after transferring from the fastest growth, I get clean looking, uniform plates.
I've always thought of it as a contaminant, and that sexier looking half was the clean section. But I also think I agree with Nichrome's earlier statement about lots of ugly growth actually being clean. Just look at some published papers. A lot of the time, their plates look like shit by shroomery's standards, yet they usually get good fruiting results.

Doobsker, it's weird I don't know what to think of that plate. I thought it was contaminated for sure but every transfer is clean, well one wasn't but that was me I'm sure. But for it to look like that is weird, looks contaminated .
