I think this button culture is doing great but I have only grown oysters before. So I haven't transferred yet



Azurescens transfer 1 of 3, the other two have contams, one is near the wall of the jar and the other is right next to the transfer and I am interested to see if the Azurescens will eat the contamn



This is 1 of 3 RustyWhyte starting to show some progress on day 4.

If I put a warm cup of water on the noc'd dishes.. do you think itll harm the spores I've got growing? This condensation is just annoying because I can't see clearly but don't wanna kill off the myc.  Its just a dot at the moment.. but still.


Here is what I'm currently doing that kicks ass:
Boil water, let sit with candy thermometer until temp hits 120F.  Transfer to SAB with media bottle in bath.  The water bath serves two purposes: 1) Agar temp, 2) Heat sink in the SAB....My SAB is hot AF while I pour.  (120F is to account for temp drop while prepping tub, ends up closer to 117 through the pour).  If my media is hotter than the water bath I wait until the temps equalize to 120F.  I pour 20 plates at a time.  Stack em in 2 10high stacks and cover arm holes with Papertowel.  Let sit for 1-2 days.  This reduces the mass temp swing that causes condensation.  Plus the condensation can escape the plates a bit.  After day two I prep for all transfers, remove heat sink (because it is room temp now).  I still end up with 2 plates that have condensation.  I set them aside and use last.  By the time I am all done my plates are clear as a bell. 
The hot cup on your plate won't hurt anything but the condensation will come back....you are essentially returning excess H2O to vapor...when it cools it will probably condense a little again.  No biggie as long as it doesn't run on the agar surface. 
Room temp helps too...I'm working in a mini-lab built from a 20ft shipping container.  I turn on a oil circulating heater for the pour and while it sits to keep a solid high 70's room temp.
Seems to work for me:)

Light or temp swings can cause concentric circles in the growth pattern... are your nights getting cold? Some cultures are quite sensitive to it... this isn't that bad: had to zoom in to see it... unless you're talking about the condensation? I was using saran wrap to cover my armholes while plates set up

Lol, i went through everything just the other day, I'm down to 9 cultures in cold storage: some of these plates are transfers out of the cold cultures I kept: going to run some test boxes to see if they're worthy, pick a winner for a few I have doubles of

RW is taking off. It is considerably more β€˜whispy’ than what I am accustomed to. Too early to start transfer? This is 7g PDA:400mL H2O:100mL grain water to get them accustomed to grain jars. The red is two drops good coloring to identify this mixture.

Disregard any grey shading, it is from the natural backlighting in my shop. The plate is all white without contam to visual inspection.

Attached is a pic of one of my petris.  It’s 11 days out, made from spore syringe drop.  And I didn’t do the whole lightning bolt move with a loop.  Mainly because I forgot. 

Look ok?

I did same drop with my own MS syringe on 6 petris about 3-4 weeks ago.  2 are growing out just fine.  Similar actually to the one I just did and pictured below.

Question:
For the 4 petris from weeks ago that nothing has happened in, and no contams at all.  Can I reuse those plates for some cloning?  I may just end up trying it, but wanted to see if y’all have ever reused petris that didn’t grow out anything and remained clean.

Is this safe to open in my house, in a SAB to get a transfer out?

Have to forgive the blob of agar. This was only of my very first transfers and I cut away to much agar from original plate.

HH, IME, luecistic varieties including RW stay fairly tomentose... might force some rhizo with less nutes, but these types of cultures have fruited just fine

Somfungi, looks good from what I can tell, id still transfer from it: I have reused plates that failed to grow anything first try without issue... breifly eyeball it open before using

NTM, whatever it is,  was already in your house, as it fell on your plate inside your house... if you've no other choice,  id transfer from as far away from it as possible, and make 2 additional transfers from subsequent plates as soon as growth leaps off far enough to avoid the donor sample... this mold has sporelated and will likely show up in at least the next plate: but they still have to germinate, so if you work quickly and cleanly, should bee able to get away from it in few plates... if you've more spores, I'd streak another just in case, watch both cultures in tandem, something will work out and you could potentially save yourself a few days/ a week

Finally have a nice culture of some Ps. Cyanescens.  My first attempts were using a MS and I was never able to get rhizomorphic growth.  Got it first try with the print I traded for.:-)



Also have some more clone plates going from the last 38g Roatan that are looking pretty wicked.

This is one of them:


Transferred a few mini pins from this old plate of Brazilians.


Have a few more RW plates germinating to try for a more rhizomorphic phenotype.


And a couple of new strains for me, GT’s and B+ have started germinating.  Also have BPK, Hualta, Golden Mammoth, KSSS and a couple others going.

Rhizo is pretty, but I don't worry about it much... tomentose cultures perform just fine IME :shrug:

Hi guys,

I'm all new to agar and to the hobby in general. But I ordered these pre poured plates and put some Paneolus tissue on there. Now, less than 48 hours later we see some growth. Rt is between 65f and 70f



Id this initial growth looking any good, or is it too early to tell?

It's bacterial, give it a few days to grow out, avoid tilting the plate in case there's excess moisture, which will easily spread the infection... it's not uncommon, and much easier to get away from than mold... I don't see any mycelium yet, the sample Wil get fuzzy, and it will grow out from there

Just parafilm, but green. My gf works as a florist. They have it lying around.

edited my response, also, try and take as little tissue as you can, IME they recover much quicker, and are less prone to bacteria

All right thanks again. Yes I didn't do that quite right. I did 5 petris with tissue. I'll post pics of them all in another 24hrs. They are my first ever though. Got the feeling there's gonna be a lot more!
