The ms plate was probably done around 1/5 or so. I just let it grow out. Got some nice thick rhizo strands coming from it. I can see how it got it’s name Creeper.

Just creeped right up.


Not sure if anyone can see any visible contams or bacteria. I use a small speck of spores in the middle of my germ plate and let it grow out from there.

Edit:
Once you do transfers it definetly speeds up. There’s already live myc and it spreads like wildfire compared to waiting for spores to germinate. Cloning is quicker as well.

thank you, thats what i was after ! interesting also that rhizos can come out later i thought it was one or the other right away

that put my mind at ease....patience is hard at first tho i grew a bunch a few years ago and had to stop bc i had no place to grow, my patience is allot better than it was when i first started tho.

It can be one or the other. Some people like using tomentose growth, and others go for the rhizomorphic. I found it best to wait and see what happens. That’s just me though.

Ps: If you haven’t already, get some new plates going, try testing some other nutrients out too.

Yes I do. It’s pretty cloudy because I use BRF. Lowered my food coloring a bit this round.

My guess is that it will be substantially cheaper to pour your own over time (the initial investment may be higher), and it will be more hassle-free with better sterile technique. BUT! I have no idea, the Netherlands are pretty foreign to me.

Bit late with my reply but thank you so much guys! I'll be attempting my first clones today! They are Copelandia (from a 100% mycelium kit) wouldn't even have tried to grow these yet, if it weren't that the hard work had already been done:)

I'll also print a bunch of Copelandia caps. So IM me if you're interested in one of those. Strain is called Ogre Boy

I'm kinda new to agar, this is my second transfer from ms(the piece is a lil big i know:).

  How small of a piece should I take to try and get some isolates to more plates? Like a certain size piece? Or literally try to grab one strand?
    Or is it something that will take many transfers to get a true isolate?

I’m not 100% on isolates. I usually take a small square the size of a single LEGO piece. You know the tiny ones? Sometimes I grab bigger ones. The smaller you get the more isolated it is I think. Not sure how that works myself though... maybe someone else could fill us in or one of us can look it up.

Its all good brother I have just heard conflicting info on it, ill do a few more transfers of different sizes and myc types to see what i can get and go from there:)  not gonna lie, I never expected agar to be so fun! Idk I ever did appreciate syringes

The smaller the piece, the better. Targeting the best looking growth, and by grabbing smaller piece, you get less genetics. When isolating, you should keep putting your isolates to fruit to see if they preform the way you want. Just cause it looks awesome on agar doesn't mean it will preform the same in a grow. It took me like six months to get a great b+ isolate. But when you have a great culture that performs well all around then you slant that bitch and do spore prints from your isolate grows. Then you will have a great isolate, and MS of that culture with the prints.

Thats what I thought, but I saw something where I guess some people do like 2 or 3+ "isolate" transfers(getrabbing smaller and smaller pieces each time) before they can actually grab individual stands.

    Like if I took the most rizomorphic section from an ms plate, transferred it and got this, can I start separating the smallest pieces of individual stands now? And put each tiny piece to a new plate? Or do I have to do this again and grab after my small piece has been moved and grown from I can take the stands from there with them actually being "pure"? I just don't want to think I have an isolated when its actually like 2, maybe more genetic lines.

i guess what im asking is: Will a single rizo-strand of this be pure? Or am I not there yet?

First transfert from SP to agar, it already look like a mono culture :shocked:



That one with a contaminant started rhizo then turn back to tomentose... :shrug:



Also, i found out that shaking agar prior to pouring will make bubbles in it, now i know why some people plates look like that, that was the first and the last time i did it.

I would bet a single strand of rhizo still contains lots of genetics. The main goal of isolating is to find a culture that performs well at fruiting. IMO, putting cultures to fruits is the most important step of isolating. Not all varieties display total rhizo growth, and just because something looks sweet on agar doesn't mean that's how it will preform in the latter stages. My best advice is, transfer from germ plate till it's clean, then fruit it, if you like it, take prints, and clones. While that's going you can further isolate the original culture, and put to fruits to see if you can find a better performing culture. Once you got a great all around performing culture, save it.

Nah that makes perfect sense! Im just gonna do 2 plates from tiny pieces of this plate, and when those are grown out I'll throw a piece of this

And each of the two further plates from this to their own jars and see what happens from each, and keep the plates in the fridge till I see what I want to continue.

I know good looking myc doesnt always mean good beyond and into fruits, its just easier to see the individual sections compared to tomotose.

Just took some tissue samples from the PM fruit, even put the cap on a plate for shits and giggles.



Did a couple RW transfers, and a couple creeper transfers as well.

I used some plates that I poured three weeks ago to germinate some spores.
Will they be too dry at this point? I sacrificed one to check it with my fingers and it still had moisture in it but was pretty hard. should I pour fresh plates and re inoculate?

They were left in my sab w parafilm. I know I can store them in the fridge but I thought I'd use them before they dried out.
