Possible contamination?

On 12/8/18 I inoculated 24 brf jars. So far it has only been 4 days.

Its directly under the inoculation sites. Im seeing a rusty or pale orangish color. Doesnt look much different from the substrate.

Are these jars contaminated with some kind of bacterial presence? 8/24 have this coloring at the inoc sites.

Looks like bacteria..did you flame your needle between each inoculation etc?

Yes :eek: I didnt flame the needle between each inoc point. But I did Flame sterilize between each jar.

Well you have possible contam points each time you touch the jar lid as you pass through..if the needle is hot each time then it self sterilises as it passes through, so its worth doing..as it only occurred on 8/24 then I'd say this was more likely than your syringe being contaminated

Im not sure about it being a contaminant from the way I inoculate. I follow a repetitive sterile technique. I took a peek at the same jars today and its looks as if that pale rust part is now growing a thin layer of mycelium.

Well if you aren't flaming between each point then you aren't practising sterile technique..its as simple as that

I have another two 12 jar batches, I inoculated following the same tek, they are all growing flawlessly. Also all the PF tek threads I've previously read, only say to flame sterilize between each jar and that to flame your needle between each inoc hole isnt needed.

Irrespective of what's written, if you think touching a needle to a lid that's exposed to bacteria and spores floating around in the air is sterile carry on. I can tell you that is not sterile. That rusty colour will be bacteria, and as your jars will have been sterile after boiling..there is only one way in

Its all done in a SAB, in a smaller clean room. As this isnt my first time I know about spores being virtually everywhere. So I make sure to wipe down the SAB, the counter, all my tools and everything ill be touching with alcohol. I also used homemade spore syringes to inoculate the jars, I was thinking a contaminated syringe could of caused it. I use the same PF Tek over and over, how would I know to flame sterilize between each hole if its not writtin in the Tek? I was hoping to get more than one opinion as to avoid this back and forth we got going. Im wondering if since mycelium is now growing over these pale spots if they will do fine.

Syringes are a known vector of contams, which is why you should move on to agar if you're serious about the hobby, all you need is a pressure cooker and some PP5 dishes..However if your syringe is then I would have expected all of your jars to be the same, but if only 8 out of 24 are, then it suggests its not the contents

You learn by experience in this hobby, and I'm just relaying mine to you based on success and failures over the last five years or so:thumbup: If its bacteria then its quite likely your jars will stall before 100%, so just give them time and see what happens. Failures are part of the process, and everyone gets a certain % of jars an bags usually, even taking all of the precautions

Here's what im seeing another day later. First 4 photos are the jars which had the pale orangish color. The last photo is a jar from a healthy batch I have going with the same tek.

Yea 100% percent a contam on those jars, you can smell it at FAE holes.
