Print, syringe, or clone? Best overall

I’m starting to work a lot more with agar after some recent fails with ms to grains, and I’m curious.

All things considered, contam rate, quickest growth, best results, what does everyone prefer to transfer to agar?

Spores from a print, a clone, or a transfer from another plate. :shrug:

Clones have the highest tam rate. Out of 5 clones I've done only 1 didn't have visible bacterial snot by the tissue. On the flip side you got a good idea what you are going to get out of a clone. The least contaminate prone clones are from invitro pins in a petri dish or container.

I no longer desire syringes and prefer prints overall. They are easier to work with, no extra water to favor bacteria and so fourth.

Overall contam rate mostly comes down to your sterile tek whether it is in a SAB or in front of a flow hood. You'll be doing a couple transfers anyways no matter if it's a print, clone or syringe to insure you got a clean culture.

I believe from what I've seen here most do prints and clones and less use syringes.

In all it will take practice and training your eye. The biggest learning curve is identifying bacteria that is riding on your mycelium. I still do not have full confidence on finding riding bacteria myself but have seen that I have improved.
The snot blobs of bacteria and yeast are easy to identify. ( A agar test of a liquid inoculant the larger pink blobs are the contams and saved me from wasting grains and coir.)



Overall everyone should do agar to save and reduce future heart aches rather than contemplate over a print clone or syringe. :cookiemonster:

So really no one way is better, it comes down to sterile technique? However prints do sound like lowest contam risk, and I’d think you get a lot more transfers from them as well.

I have some clones on agar now and hopefully at least on is viable, probably should’ve made some prints too huh

I don't have issues with clones... In fact, I've used tissue plates from bacterial tubs for A2G, how are you taking yours RIP? Cutting the fruit open? Sounds like cross contamination from outside to inner tissue... taking prints when possible is always a good idea: I clone and print the same fruit sometimes lol

You can transfer from any culture many many times...

Unless you're growing in a sterile lab, spore prints/syringes will never be free of contaminations. I don't see why you'd have worse contamination rates with clones.

I agree. Though I haven't tried proper isolation yet. It seems cumbersome to search for good genetics by fruiting every possible isolation.

I like isolating from clone culture... you know it'll fruit, and narrows things down substantially than isolating from spore

You probably want to just take some prints, as well as clone a couple of the larger fruits, from the faster growing clusters. Its not like you have to choose one or the other

I would say probably cloning has the best results if I had to pick one. :shrug:
