Agar plates never took off

Used spores to inoculate around 15 plates of agar and none of them have shown growth 26 days later, and I'm wondering what I might have done wrong for this to happen. I know they could still show growth but at this point I'm just going to assume they won't and start trying to fix my process.

I followed bod's agar guide and used the 2% mix for making my MEA. I used telephone brand agar and muntons light spraymalt. Nothing grew on these plates no contams or anything. I did however touch one uninoculated plate and then isolated it to see if it would contam, which it did, so this shows that at least something could grow on the agar.

I'm not really sure what went wrong but I'm thinking about trying a different recipe for agar maybe moving away from malt extract. I also won't be using spores.

Anyone else have any suggestions?

Your agar mix is fine.  How old are the spores?  They do not get more viable with age.

The syringe was made in mid September, so definitely not old. There were also tons of spores in the syringe. I wonder if the malt extract I used just isn't good for agar?

I used 1 drop per plate, maybe this wasn't enough but I can't imagine 15 plates not getting enough spores from 1 drop since that's normally the recommended amount.

I'm assuming the print you made the syringe from is new as well? One drop per plate is definitely enough.  Where did you acquire your malt extract from?  Can't say I've heard of spraymalt before. I use light malt extract from the home brewers shop. You could test the syringe on a small batch of grain jars to help troubleshoot.

I got the syringe online so no idea.

It was this stuff malt. I remember reading a post somewhere where someone said spraymalt was good, havent heard much about it anywhere else though.

I'm thinking about just trying different recipes for agar, or maybe if I can get my hands on some malt extract that is known to work then I'll try that.

What malt extract do you use?

Couldn't tell you a brand name of the ME.  It's just whatever my local brewing shop had, which was a light dried malt extract.  That spraymalt is expensive.  I bought a pound of the aforementioned ME for $4.  Works great.  I would say in the future to work with prints instead of syringes.  I don't know why, but I always have much better results inoculating agar with spores from a print.  This isn't always possible, like with varieties like PE.  My work around for that was inoculating a small batch of grain jars with the syringe, and then transfering the colonized grains to agar.

Admittedly, I am a noob but I had a similar experience but had a few other reference points so I can pretty confidently say that the spores are very old or in some other way not good so that is what I would suspect. 

I used spores from a fresh prints I had made on two plates.  Those were off and going in less than 36 hours.  Only one plate out of 12 inoculated from 3 different syringes germinated, and I used a lot more than 1 drop. With this info I can confidently pin it on the spores because all of the plates were made at the same time.  Oh well, when those fruit I will do a spore print and have capture the strain.

Now a slightly off topic but related question.  IF the spores have not germinated and the plates are not contaminated can you used the plate with more spores from the syringes?

Technically yes, but I wouldn't consider it best practice.  Give it a whirl.:shrug:

I'd just pour it new. This hobby taught me that you shouldn't leave anything to chance if you don't have to. Or unless you don't mind wasting days or weeks
