Agar to shoeboxes: a little lost, need a roadmap

Hi there

I recently inoculated 10 x no-pour agar plates from a spore print. I lost 2 to contamination and so have 8 that are looking good and clean.

I'm aiming to end up fruiting maybe 4 to 6 shoeboxes, so am trying to work backwards in terms of quantities: number of plates and number of jars.

I'm ready to do my next transfer (agar to agar)  and have 10 fresh no-pour plates ready to inoculate from my initial batch.

This is where I get lost.

1. Should I transfer a wedge from each inoculated plate to another clean plate, so I end up with 8 new plates? Or, do I only transfer some of my current 8 plates? Should I aim to inoculate more than one receiver plate from a single donor plate?

2. Do I simply discard the donor plates after the transfer? Keep them in a fridge as a back up?

3. I'm guessing I need to end up with say, 12 x quart grain jars (allowing 2 per shoebox for up to 6 shoeboxes). How many plates of clean mycelium should I be aiming for in order to inoculate 12 jars (quart sized)? Should I just aim to tiger drop and therefore one plate per jar of grain?

4. How long can I keep inoculated plates in the fridge if I am not going to need them for this fruiting run?

5. How focused should I be at this stage on isolating a strain?

i usually take 5 xfers per agar jar ya i throw away the plate i just used for the xfers to refil with fresh agar
i use a half of jar per jar so 5 agar jars per 10 qts of spawn i like to keep shit on the 5 jar thing

Don't isolate a strain imo if you don't have mushrooms already.

They can sit in the fridge for quite some time. I've been sitting on my very first set of agar for months and months ...

Don't need them often going g2g.

Transferring agae the way I do it is draw a pizza and end up with like 8 slices and use those for transfer / inoculation.

I transfer one pizza slice to one new agar, or one pizza slice to a quart jar.

Really you need 1 clean agar and you can inoculate (I do 7 because that's how many quart jars I fit and prep in one run)

Then you just need one clean jar to go g2g to another run, or 7 more quart jars.

Then you can g2g this jars as well.

So if say 5/7 of the jars you inoculate with 1 agar plate take and are clean, you can with up with 35 (give or take for contams) clean jars

That's how I would do it. I would try one agar plate to 7 jars at a time and if they all fail try again (they won't unless you're fucking up)

And g2g the ones that colonize clean

Oh I forgot to add

With agar I just drop one pizza slice right in the middle of the new dish agar.

With quart jars you can use more pizza slices, and more slices per jar, potentially, and speed up colonization rate a bit, potentially.

But imo it just seemed more sensible to get a full run out of one agar dish since they colonize fast anyway then you have more jars to g2g with :shrug:
