I'm glad you found a working additive for T-Gel, I am still experimenting with various additives.

Growing Tarragon horizontally as in from trays? -- Yes.

The MG works, but adds no carbon, the gelatin recipe has ended up the better version so far, it has better but slower growth. < Still tweaking.

Yes of coarse be aseptic for proper work, I am 'testing' various materials and open air assembly.

Thanks, T-gel is a cool concept :laugh:

I checked and malt contains lower / free sugars like glucose as well as oligosaccharides called maltodextrins. If you can substitute for the other various micro-nutrients well enough it may be worth checking the difference between malt extract and pure maltodextrin.

Am just wondering what the real differences are in macro-nutrient uptake between bacteria and our fungal cultures and how to maximize that. May be good to use as much selective nutrients like higher carbohydrates or materials composed mostly of lignins?

And I guess you can apply a spectrum of different T-gels depending on whether you want selectivity for your lignocolous fungus or instead a bit more rapid growth? For rapid growth you would allow for small amounts of easily digestible sugars etc even though bacteria could utilize them?

Indeed it's pretty fun to work with. The tea is essentially selective to laccase capable organisms.
I am also looking at other 'resistant' materials, that don't germinate aerial invaders.

Currently I am trialing agar with gelatin as the only additive, no tea.

Update: Tarragon oyster detects and utilizes animal proteins for both carbon and nitrogen-other.
I should have added a black dye to the media, as my surface is white, visibility is low.

Hopefully the media won't turn to a liquid due to the agar base.

The protease enzymes still function at pH 8.0.

Standard assembly turns to a liquid, the agar falls and sets at the bottom.
The gelatin sits on top and hardens much later, in a fridge.

Using gelatin with T-Gel works as normal.

The best recipe's are recipe 6 then 2, after 4-5 days.

----

Recipe 1: Added 2g sucrose and 0.1g MG, per 100g extract.  < Got mold after 4-5 days, healthy.
Recipe 2: Added 2g 240B gelatin, per 100g extract.  < Slow for 4-5 days, medium speed, very populated, healthy.

Recipe 3: Added 1.6g of sucrose, per 100g extract.  < Not much change, medium weak growth, healthy.
Recipe 4: Added 0.1g MG, per 100g extract.  < Slightly slower, very populated, healthy.

Recipe 5: Added 0.2g MG, per 100g extract.  < Negative impact to health and speed, needs more carbon.
Recipe 6: Added 2g gelatin and 0.2g MG, per 100g extract.  < Very slow, slightly odd for 4-5 days then very healthy.

No contamination germinated with added gelatin, MG, or gelatin and MG.
Too much MG without an added carbon source causes a problem.

Suggested recipe for gelatin and T-Gel:

100g > Water  |  2.4g > Agar Powder  |  1.6g > Black Tea |  0.8g > 240B Gelatin (See Here).


I used the microwave oxidization assembly method, it sets as normal.

Add the gelatin after the cook, stir and reheat for 20 seconds.
The media will change from lightly cloudy to normal.

Tip: Reheat the media when it's hand cool.

Next test.

Recipe 7: Added 0.8g gelatin and 0.08g MG, per 100g extract.

I'm ordering a new culture of Tarragon from the only vendor I can find (same one).
This time it will be taken directly to T-Gel, for cleaning (not clean).

I'm refraining from sending new transfers until then.

The new dowels should turn up this weekend. This time I did not order the rye spawn, hoping the dowels are better.
Previously I used the spawn on a substrate expecting normal results, then taking a transfer after.

At the time I was using seaweed as an additive, which contains additional bacteria.

T-Gel succesfully cleaned the visible yeast-bacteria combo, but not the added seaweed bacteria.
I believe the added bacteria is Archaea, which can produce laccase and grow on T-Gel.

The Archaea-other, produces dark patching on T-Gel and WL-Tek, hard to see.

I'm hoping the vendor dowels will be free from laccase invaders.

I am impressed. Tarragon dowels seemed to be axenic. purchased 2 years ago.
Some observations: Tarragon oyster grew huge kombucha like blobs in liquid culture stored at 2 degrees Celsius. But substrates fruit after exposure to temperatures bellow 10(?) Celsius at any stage of incubation. Temperature is an approximation. Incubation is greatly accelerated at temperatures above 20 Celsius but I have never incubated it above 23 so...

I always wanted to isolate some new cultures from spores but always busy, now you gave me the inspiration...

No problem, I am still annoyed I cannot find a wild one, nor another vendor. Solarity also warned me about the current supplying vendor.

The kombucha you describe is similar to what I got the first time the transfer culture was placed onto T-Gel.

You can use the wedges I sent, it could cleanup entirely, fruiting and taking spores is not an issue.
Be prepared to get visible browning, in the spawn and substrate, and some loss of yield.

Any containers will need proper sterilization after, or be disposed of.

There is not much Archaea-other on the wedges.

Source, organic liquid seaweed, can be used on vegetables:

   

Not ideal for primary "decomposers".

TravelAgency who received a pre T-Gel peg, said the fruits where unique and special.
He managed to produce good yield even without the T-Gel cleaning I used.

I believe he and another made light attempts at cleaning it.

T-Gel, T-Gel + Malt extract, Malt extract. You should see regeneration after 4-8 hours, and radial recovery after 12-18 hours.

I'm going to throw off some of your very well researched and documented findings-theories. If you are talking about lignicolous mycelium you can also add polyphenols.
If we take for example lignin as the phenol, it contains no glucose, no sugars whatsoever, however lignicolous mycelium will receive carbon and other.

T-Gel contains about 0.01g of natural sugar per 200g of media, this is insignificant, the phenols are the main source of carbon, not sugars.
The mycelium will cleave the phenols, produce solubles, absorb them, and then continue "at will" to produce [X] material(s).


T-Gel + MG (phenols + macro-micro nutrients) had a significant impact to growth.
T-Gel + sucrose (phenols + sugars) had no significant impact to growth.

This can only be explained by utilization of phenols as carbon.

Tip: Lignin is also a phenol based material

Tea leaves, Ready-to-drink.

Proteins, varies:  [ Cx | Hx | Ox | Nx ] x. -- Trace, minimal.
Polyphenols, varies:  [ C6 | H5 | O ] x. -- Phenols.
Cellulose: [ C6 | H10 | O5 ] x. -- Trace, debris.

Theobromine:  C7 | H8 | N4 | O2. -- Phenol.
Theophylline:  C7 | H8 | N4 | O2. -- Phenol.
Caffeine:  C8 | H10 | N4 | O2. -- Phenol.
Theaflavin:  C29 | H24 | O12. -- Phenol.
Tannins:  C76 | H52 | O46. -- Phenol.
Catechin:  C15 | H14 | O6. -- Phenol.

Carotene:  C40 | H56. -- Hydrocarbon.

Glycogen: [C6]--[H10]--[O5], (contains glucose).
Ergosterol: [C28]--[H44]--[O], (essential).


Cellulose: [C6]--[H10]--[O5] x z, (same as glycogen, contains glucose).
Starch: [C6]--[H10]--[O5] x z, (same as glycogen, contains glucose).
Phenols: [C6]--[H5]--[O] x z, (phenols vary, not glucose based).
Sugar: [Cz]--[H2]z--[Oz], (sugar varies, ideally use glucose).

Protein: [Cz]--[Hz]--[Nz]--[Oz] x z, (contains both carbon and nitrogen).


z = Number of total units linked.

Enzymes

Disclaimer: Optimal pH varies with the organism, and it's enzymes.

Cellulase (cellulose): 4.5-8.5.
Laccase (phenols): 5.5-6.0.
Amylase (starch): 3.0-7.0.

Protease (protein): 2.5-11.


For cellulase there are four types:

Alkaline cellulase: 7.2-8.5.
Neutral cellulase: 6.0-8.0.
Hybrid cellulase: 4.5-7.0.
Acid cellulase: 4.5-5.0.


For amylase there are three types:

α-Amylase: 6.7-7.0.
β-Amylase: 4.0-5.0.
γ-Amylase: 3.0.


Laccase remains stable at pH 5.0-10.0.


For protease there are three types:

Alkaline protease: 7.0-11.0.
Neutral protease: 6.0-9.0.
Acid protease: 2.5-4.0.

The vendor has apologized for the delay, most likely making a fresh batch. While I wait for that I decided to try 10% bleach peg's as transfer tools.
10% bleach will kill-inhibit bacteria-yeast-archaea, so it should kill-prevent the archaea that I currently have as an invader organism.

The multi-cellular mycelium can distribute materials to and from the peg, allowing it to tolerate the 10% bleach.
Unicellular organisms should die on contact, or be restricted, the pegs also provide an airy surface.

Here are some images of the pegs 2.5 days after placement, a little slow but working.

           

The mycelium appears to be thicker and more populated around the pegs.

Crazy how fast this culture colonizes. Just put a wedge off clean agar onto grains 2 days ago, and it's already 30-40% colonized. Insane!
