Tarragon Oyster T-Gel Wedges [Giveaway]

I have [5x] T-Gel wedges of the rare Tarragon Oyster that I am willing to send out at the end of next month.
There will be more wedges soon, and I will update this thread when there is more to send.

Tarragon Oyster (Pleurotus euosmus):


Tarragon Oyster Mushroom is a very rare oyster mushroom which is thought to be only found in the British Isles and is not cultivated commercially at all.


Mycelial Characteristics: White, longitudinally linear, cottony aerial fast growing and classically oysteresque. Soon after colonizing a petri dish of MYPA, the mycelium tears off in thick sheets.

Microscopic Features: This mushroom produces pale pinkish/lilac spores.

Suggested Agar Culture Media: Malt Yeast Agar (MYA), Malt Yeast Peptone Agar (MYPA), Potato Dextrose Yeast Peptone (PDYA), or Oatmeal Yeast Agar (OMYA).

Spawn Media: Grain spawn for the first two generations, hardwood sawdust spawn for the final stage.

Substrates for Fruiting: Hardwood sawdust, cereal straw, cottonseed hulls, sugar cane bagasse, coffee wastes, paper products, and many other materials. This mushroom will probably grow on many more substrate materials give modest experimentation.

Yield Potentials: 75-100% biological efficiency, greatly affected by the size of the mushrooms at harvest, and the number of flushes allowed.

Relative Humidity: 95-100%
Duration: 7-10 days
CO2: < 2000 ppm
Fresh Air Exchanges: 4-8 per hour
Light Requirements: 750-1500 lux


Fruitbody Development:

Temperature: 70-80° F (21-27° C)
Relative Humidity: 90-95%
Duration: 4-8 days
CO2: < 1000 ppm
Fresh Air Exchanges: 4-8 per hour
Light Requirements: 750-1500 lux


Cropping Cycle:

3 crops, two weeks apart.


Comments: With a distinctive and delicate trumpet shaped cap, as it’s name suggests, it’s smell and taste resembles the herb tarragon.

Extra data for Tarragon oyster:

Active enzymes (general): Cellulase (cellulose), Laccase (phenol's), Amylase (starch), Protease (protein) | Sugars (such as Sucrase).

Carbon sources (general): Cellulose, Starches, Sugars, Proteins, Phenol's, Other.
Nitrogen sources (general): Protein, Some Phenol's, Ammoniacal, Ureic, Other.

Tip: Tarragon doesn't like too much acidity, or too much phenol's.

Tested to work at pH 6.0-8.0, I've not tried below 6.0.

The optimal pH with paper was 7-7.5.

Results of Pine v Hardwood Pellets on Tarragon Oyster

Pine took an early lead and stayed that way, getting to 100% 6 days before HWFP, and has already pinned by the time I put the Hardwood in the FC, and also colonized much thicker. Yields were also in favor of the Pine by several ounces.

Colonization differences:



Pine:








HWFP:





Recipe was 60 cups pellets, 20 cups wheat bran, 5 gallons water for both. Harvest on Pine was a full week ahead of Hardwood.

Conclusion: Patterson’s Horse Bedding Pine Pellets colonize faster and produce Higher yields than do HWFP from Tractor Supply.

Caveat: I have used better quality HWFP in the past- there are a lot of small chunks that don’t turn into pure sawdust which may be a factor.

Observations: Tarragon is very stemmy, I had Golds and Bratslavian growing along with it that were far less stemmy than the Tarragon. It is also a beautiful strain, a grayish purple with dark purple stripes on the stipe and a speckled lighter purple for the caps. Haven’t cooked with them yet so I can’t attest to taste yet.

That's some nice BE you got there, not sure what's causing the wavy caps.
Excellent write up and description, are you collecting spores?

You can clone new fruiting genetics that suit more.
The vendor cloned a horizontal fruit.

I can share agar of this if it's so popular, in a thread i will make for that, as soon as I confirm that the discoloration and slight slickness I see are just related to laccase on T-gel and not bacteria or anything like that. You may need to reimburse shipping though if it's gonna be a lot of wedges cause I have no income... don't worry mail is quite affordable here relatively to certain countries (still it adds up)

Is this species like Hericium in that FAE / CO2 level is supposed to be a serious factor in how strong the aroma / taste is?

How might one get too much polyphenols in the substrate?

It should be laccase enzymes (brown-tan). Currently I am adding tea, which adds acidic phenol's.
Adding too much tea slowed down the mycelium, wood can also be too rich.

I will do more wedges soon on the updated T-Gel, I am using the microwave assembly, and adding my additives after the cook.
Currently I have 3 plates made on different days, a visual comparison is being made, on population and health.

Microwaving the pre-extract causes it oxidize, in a similar fashion to laccase enzymes.
The conversion also causes to agar to set very hard, without any sugars.

Recipe 1: Added 2g sucrose and 0.1g MG, per 100g extract.
Recipe 2: Added 2g 240B gelatin, per 100g extract.

T-Gel Recipe 1 has germinated and grown out a random mycelium after only 4.5 days.
Recipe 2 is yet to contaminate and is producing growth equal to Recipe 1.

Bottom: Control, Middle: Recipe 1, Top: Recipe 2.

It's lignicolous mold, there is a green-blue patch appearing. It's also penetrative (see below, right side - internal white).

Surface growth is more populated and aerial with Recipe 1 and Recipe 2, very similar results.
The both samples have penetrative growth, through the wedge and the main agar.

Recipe 1, is 2.5 days ahead of Recipe 2 in terms of sample assembly.

Some data and notes for cultivators:

The T-Gel plate with added MG, but no sucrose, did not contaminate from open air, while with added sucrose did, rapidly.
Gelatin (change to 100g) is about 85-95% proteins, nutritional proteins count both as carbon and nitrogen.

Recipe 1, is producing equal results to Recipe 2 due to the addition of the sucrose as carbon.
In short the mixture of both carbon and nitrogen acts like adding proteins, in a way.

Tarragon oyster will also utilize other carbon sources with MG (WL-Tek).
It decay's the cellulose as carbon (no sugar, no starch).

   

Use clean spawn, wait 2-3 weeks.

These 3 plates where poured open to air, and set open to air, that's about 2 hours.

The lignicolous mold is very healthy, very populated and well fed.
Cubensis also germinates on sucrose + MG, in 4 days.

So t gel made with MG and no added sucrose, was poured hot in a dish in open air, allowed to set in open air approximately 2 hours later, and to this date it still hasnt germinated any airborne spores/bacteria/yeast?

How long ago did you pour? The other plate with sucrose that contaminated quickly, it was poured at the same time?

Not quite that much of a test, I am testing open to air assembly, with the push of setting the agar open to air.
All samples are inoculated with an oyster sample after fully setting, general data is recorded.

I am testing effects on growth, and chance to contaminate from assembly-other.
So far Recipe 2 is slower than Recipe 1, Recipe 1 has only mold.

All 3 samples are clear of visible bacteria-yeast.

Recipe 4: Improved population, no contamination.

Thanks, Golden oyster loves: Sunlight + water (rain) + high humidity + high O2.
Tarragon oyster needs far less light, but it produces thicker stems.

Tarragon oyster will also fruit very well horizontally.

Recipe 5: Added 0.2g MG, per 100g extract:

Improved laccase production (more browning), I believe this is due to two reasons, enzymes are proteins, they are composed of C-H-N-O.
Laccase also contains copper, so it's C-H-N-O-Cu, the MG adds nitrogen and copper to the T-Gel media, which contains C-H-O.

In general laccase will produce C-H-O from phenol's, and the N-Cu from the added MG, more enzymes = more C-H-O.

Recipe 2, 4 and 5 produced an increase to population rate, thickness and overall growth potential.
The slowest of the three is Recipe 2, nutritional proteins provide C-H-O and N-Other.

I don't have malt extract so tweaked the T-gel recipe/concept a little before (and have in the process also created agar on which nothing seems to grow, lol), got a good one now though:

Around 150 ml water, simmer in it about 5-6g oats and a dash of bran, extract a teabag of black organic tea in it and strain carefully into erlenmeyer with 4(+) g agar powder suspended in about 50 ml cold / RT water, to make 200 ml agar.

I was already dying my medium for contrast - now that I have a few more colors I can color code for different media... although T-gel does this spontaneously by being brown. :wink:

I suspect that maltodextrine may be a suitable replacement for the oats or malt extract but not sure (I think oligosaccharides are mostly the point of the malt), but if I were to purchase new ingredients might as well go for the malt extract. Using yeast nutrition can provide micro-nutrients - mostly minerals and some other inorganics etc - bran surely has a richer organic micro-nutrient mix like including protein.

I was having a lot of tomentose growth on my PDA, and while growth wasn't necessarily slow it tended to be wispy initially (later consolidated), I believe I was allowing too much potato starch to be dissolved / suspended.

On the tweaked T-gel i mentioned I now am getting much more beautiful full white growth (that is, with Ovoids, subs, and now also gyms and Ps Mex though I have no idea how lignicolous it is as nobody grows truffles on sawdust lol) and it should work for gourmet woodlovers as well.

Might have to check differences between MG, bran and yeast nutrient (nutrivit)..

By the way: while testing open air conditions might be interesting in *some* ways it seems like a bad plan because contaminants can still hitch a ride secretly even if they are not encouraged to grow and become dominant. Would you not expect problems once you go to spawn and bulk? Or are you just experimenting to see more acute sensitivity to seriously contaminate?
Just saying: you might wanna point out that you don't recommend open air conditions at this stage even with actual T-gel use if it is not meant for that.

I'm glad you found a working additive for T-Gel, I am still experimenting with various additives.

Growing Tarragon horizontally as in from trays? -- Yes.

The MG works, but adds no carbon, the gelatin recipe has ended up the better version so far, it has better but slower growth. < Still tweaking.

Yes of coarse be aseptic for proper work, I am 'testing' various materials and open air assembly.

I checked and malt contains lower / free sugars like glucose as well as oligosaccharides called maltodextrins. If you can substitute for the other various micro-nutrients well enough it may be worth checking the difference between malt extract and pure maltodextrin.

Am just wondering what the real differences are in macro-nutrient uptake between bacteria and our fungal cultures and how to maximize that. May be good to use as much selective nutrients like higher carbohydrates or materials composed mostly of lignins?

And I guess you can apply a spectrum of different T-gels depending on whether you want selectivity for your lignocolous fungus or instead a bit more rapid growth? For rapid growth you would allow for small amounts of easily digestible sugars etc even though bacteria could utilize them?

Indeed it's pretty fun to work with. The tea is essentially selective to laccase capable organisms.
I am also looking at other 'resistant' materials, that don't germinate aerial invaders.

Currently I am trialing agar with gelatin as the only additive, no tea.

Update: Tarragon oyster detects and utilizes animal proteins for both carbon and nitrogen-other.
I should have added a black dye to the media, as my surface is white, visibility is low.

Hopefully the media won't turn to a liquid due to the agar base.

The protease enzymes still function at pH 8.0.

Standard assembly turns to a liquid, the agar falls and sets at the bottom.
The gelatin sits on top and hardens much later, in a fridge.

Using gelatin with T-Gel works as normal.

The best recipe's are recipe 6 then 2, after 4-5 days.

----

Recipe 1: Added 2g sucrose and 0.1g MG, per 100g extract.  < Got mold after 4-5 days, healthy.
Recipe 2: Added 2g 240B gelatin, per 100g extract.  < Slow for 4-5 days, medium speed, very populated, healthy.

Recipe 3: Added 1.6g of sucrose, per 100g extract.  < Not much change, medium weak growth, healthy.
Recipe 4: Added 0.1g MG, per 100g extract.  < Slightly slower, very populated, healthy.

Recipe 5: Added 0.2g MG, per 100g extract.  < Negative impact to health and speed, needs more carbon.
Recipe 6: Added 2g gelatin and 0.2g MG, per 100g extract.  < Very slow, slightly odd for 4-5 days then very healthy.

No contamination germinated with added gelatin, MG, or gelatin and MG.
Too much MG without an added carbon source causes a problem.

Suggested recipe for gelatin and T-Gel:

100g > Water  |  2.4g > Agar Powder  |  1.6g > Black Tea |  0.8g > 240B Gelatin (See Here).


I used the microwave oxidization assembly method, it sets as normal.

Add the gelatin after the cook, stir and reheat for 20 seconds.
The media will change from lightly cloudy to normal.

Tip: Reheat the media when it's hand cool.

Recipe 7: Added 0.8g gelatin and 0.08g MG, per 100g extract.

The new dowels should turn up this weekend. This time I did not order the rye spawn, hoping the dowels are better.
Previously I used the spawn on a substrate expecting normal results, then taking a transfer after.

At the time I was using seaweed as an additive, which contains additional bacteria.

T-Gel succesfully cleaned the visible yeast-bacteria combo, but not the added seaweed bacteria.
I believe the added bacteria is Archaea, which can produce laccase and grow on T-Gel.

The Archaea-other, produces dark patching on T-Gel and WL-Tek, hard to see.

I'm hoping the vendor dowels will be free from laccase invaders.

I am impressed. Tarragon dowels seemed to be axenic. purchased 2 years ago.
Some observations: Tarragon oyster grew huge kombucha like blobs in liquid culture stored at 2 degrees Celsius. But substrates fruit after exposure to temperatures bellow 10(?) Celsius at any stage of incubation. Temperature is an approximation. Incubation is greatly accelerated at temperatures above 20 Celsius but I have never incubated it above 23 so...

I always wanted to isolate some new cultures from spores but always busy, now you gave me the inspiration...

You can use the wedges I sent, it could cleanup entirely, fruiting and taking spores is not an issue.
Be prepared to get visible browning, in the spawn and substrate, and some loss of yield.

Any containers will need proper sterilization after, or be disposed of.

There is not much Archaea-other on the wedges.

Not ideal for primary "decomposers".

TravelAgency who received a pre T-Gel peg, said the fruits where unique and special.
He managed to produce good yield even without the T-Gel cleaning I used.

I'm going to throw off some of your very well researched and documented findings-theories. If you are talking about lignicolous mycelium you can also add polyphenols.
If we take for example lignin as the phenol, it contains no glucose, no sugars whatsoever, however lignicolous mycelium will receive carbon and other.

T-Gel contains about 0.01g of natural sugar per 200g of media, this is insignificant, the phenols are the main source of carbon, not sugars.
The mycelium will cleave the phenols, produce solubles, absorb them, and then continue "at will" to produce [X] material(s).


T-Gel + MG (phenols + macro-micro nutrients) had a significant impact to growth.
T-Gel + sucrose (phenols + sugars) had no significant impact to growth.

This can only be explained by utilization of phenols as carbon.

The vendor has apologized for the delay, most likely making a fresh batch. While I wait for that I decided to try 10% bleach peg's as transfer tools.
10% bleach will kill-inhibit bacteria-yeast-archaea, so it should kill-prevent the archaea that I currently have as an invader organism.

The multi-cellular mycelium can distribute materials to and from the peg, allowing it to tolerate the 10% bleach.
Unicellular organisms should die on contact, or be restricted, the pegs also provide an airy surface.

Here are some images of the pegs 2.5 days after placement, a little slow but working.

           

The mycelium appears to be thicker and more populated around the pegs.

Well I put the wedge you sent on a plate of agar which grew out fast. Then cut a tiny wedge out of that and put in 1qt of oats. It’s about 60% colonized now without any shakes.

Ok, so glucose and glucose-fructose. The interesting thing is the King oyster I sent to mattisfat also ended up more prolific than normal.
I'm guessing the conversion of the phenols in T-Gel to essential carbon, has improved the internal BE of the mycelium.

Technically, the phenols would be converted into glucose-other, so utilization of glucose may have improved.

The real question IMO is whether the opposite works just as well. Fungi like switching substrate to some extent (though very big changes can apparently stump them temporarily I guess), could just be that variety is 'the spice of life' in that a broad spectrum of nutrition is offered.

It makes sense that including polymers of sugars and short chains or monomers in the medium is best similar to how it is easy for our bodies: have some readily available and use the rest for the slow burn.

Both glucose and fructose seem like the best sugars for fungi to me considering both are pretty easily used in chitin synthesis.

I guess you may be right Ferather, about aromatic degradation pathways still fueling the krebs cycle. And I guess this is also why at least supplementing one of these energy sources with the other should work best. In wood the potential for both is present.

I guess this suggests that you wanna incorporate a little bit of phenols in a medium otherwise based on sugars.

And yeah pretty crazy culture, it climbs the walls :laugh:

Indeed, wood naturally will be cellulose-phenol based (in general, there are more materials), about 50% carbon and 0.1% nitrogen (weak).

From reading there is also energy storage based phenols, which can contain sugar as the backbone and-or bond.
Phenols come in a large variety, simple and complex, some are acids, some contain no sugar.

Note: Proteins contain glucans-other and fixated nitrogen.

Yes it has a very fast recovery time, it takes about 4-8 hours to recover, usually.

The bleach pegs appeared to have worked as intended first time round, however once colonized I will repeat one more time.
Currently I am using T-Gel plain, no gelatin or other additives, population and rate of growth have improved.

Using a visual estimate, the mycelium has improved by 25%, growth is thick, populated and fast.
The multi-cellular mycelium tolerated the 10% bleach, there are no visible invaders.

Household bleach:

Disinfectant: Sodium Hypochlorite 4.47g per 100g.
Anionic Surfactant: Less than 5%.

Is the agar in those tubes just water and agar to keep the peg from drying out or is it some kind of media that the mycelium will grow on too?

I've been wanting to try this peg method but haven't even cut the toothpics yet.  Looks like it could be a great way to ship cultures.  If you could ship them in a small plastic bag instead of the centrifuge vial, it could make international shipping cheaper but they might dry out too fast.

If the peg is well hydrated and in a sealed container, it shouldn't dry out unless you are sending somewhere with 30 day or longer snail mail delivery. Then you can put a little agar in the bottom to keep it lively.

There is no agar in those tubes, just wood pegs with mycelium on it, the condensate is because they where kept in a fridge.
The shipping will cost the same for vials or bags, in fact, I put the vials into a zip-lock bags, extra safety.

The pegs are cut with wire cutters, you can also snap them, PC them in a small container.
You can add agar to the bottom of the vials, using folded card with holes.

The added agar will prevent rolling, touching and drying out.

If shipping is the same for vials or bags then vials would obviously be the best option.  I was thinking the little pegs could be put in a small bag (maybe an impulse sealed bag) and sent in a normal envelope.

I've never shipped anything out of the USA but assume the cost would be pretty high.  How much does it cost to ship your pegs from the UK to the USA?

I'm going to try sterilizing some pegs and see how some other oyster cultures will grow on them.

Working as intended, the mycelium visibly looks healthier. The mycelium has pushed about 15mm from the peg.
The growth is radial, so the full diameter is about 30mm, the growth has changed around the edge.

The new growth is more populated, thicker and more visible than the starter growth.

Side note:

The added gelatin improves BE by 50% in comparison to plain T-Gel.

10% bleach peg to plain T-Gel, no gelatin:

Total growth is currently 45mm in diameter, +15mm, +7.5mm center to edge, since yesterday, and still accelerating.
PSA + ammonium chloride produced a 50mm diameter after 4 days for Pleurotus ostreatus (see here).

The T-Gel plain, without additives, is relatively close in speed, but can take more nitrogen.
Adding MG nutrients or adding proteins, had an almost identical effect on BE.

10% bleach peg, WL-Tek + 10% CaCO3, trial:

Started another trial, in a similar way to the 10% bleach peg. I added 2.5g of ultra fine calcium carbonate, for 25g of paper pellets, to my water.
The mixture of CaCO3, Miracle-gro and yeast nutrients produces no side effect gasses, or anomaly's, it mixes perfectly.

If I added bleach to MG and YN, toxic gasses would be released, do not do it, literally ever!

The end pH is an average of 8.5, the unreacted CaCO3 will convert overtime.
Water retention has increased slightly, the smell is cleaner.
