Next Step After BRF TEK - LC from Tissue?

I'm trying to figure out the easiest way (for a new person) to put my current grow to use so that I don't have to buy a new syringe or more spores. I'm also trying to sort of learn one new technique at a time (which is why I'm not really looking to dive into agar just yet) so I can figure out where I go wrong if I run into contams or issues.

Right now I'm on my second flush of my first ever grow (BRF tek). Going pretty well. I think I'm going to stick with the brf next time around but thought I might try making an LC from tissue from my best growing specimens from the current crop and using that to knock up some new BRF jars.

I have a PC for sterilization.

Any tips or suggestions on starting an LC from tissue? Or am I better off just making a spore syringe and using that? I guess I could do both to be safe -- take a few prints and try the LC as well.

Basically my understanding is that you just prepare the LC as usual but instead of putting in spores, you cut (with a sterile knife) a bit of the stem tissue and toss it in. Is that right?

My best tip for making LC from tissue is not to do it. Take some spore prints and make some syringes or get into agar.

Trust me from experience tissue needs to be put to agar before using it to inoculate anything else. You need to get your clean healthy myc away from any contams it's carrying. Some people get away with tissue>lc sometimes but very low success rate. You'll waste more time and energy trying to use tissue as inoculate without cleaning it up on agar first. If your serious about this hobby just learn agar off the bat it's not hard, and it's necessary for consistent success.

Thanks Swazey, I hope OP doesn't think I'm sounding like a dick. I learned the hard way, and I'd hate to see anyone else waist the same amount of time and energy as I did on high risk methods.

Cool, thanks a lot you all for the advice!  I think I'm going to:

1) try making a spore syringe + order another strain as my go-to plan for the next crop

and then

2) look into agar and experimenting with that alongside grow #2

I'm really into the sciencey side of things (reminds me of college bio labs and stuff) but just a bit hesitant as I don't have a flowhood or incubator or anything. Also, I'm going to be moving in a few months so I don't want to get into a massive growing setup until I'm somewhere more stable.

I've been having so much fun with this stuff so far! I think I enjoy growing them almost as much as eating them :smile:

Making syringes from homemade prints is high risk as well. The syringes sold by reputable vendors are made in sterile condtions. Without a flow hood, SAB or sterile lab, printing and syringes at home isn't a good idea. The money your going to spend on prints and syringes from vendors you can get some MEA, petri dishes, and build a still airbox and begin your agar quest. I just ordered some MEA and 2 sleeves of petris and it was like $35 including shipping. Of course I already have a flow hood, but you can build a SAB for under $10. Honestly $50 and you can be up and running with agar in no time. I'd say buy another syringe, get your supplies for agar, start your next grow with the syringe from a reputable vendor. By the time you get fruits off your next grow you'll have all your supplies to clone and isolate. You'll be rewarded by inoculating with clean healthy cultures.

yea tissue to LC is dangerous.  tissue to agar to 2-3 more plates to lc would be better.  i still fuck with agar>lc and still get like a 30% contam rate.

I just assumed he didn't have a SAB or flow hood. That's why I said without one of those two items it's high risk. I know I sound like an agar Nazi but if I could give only one word of advice to new growers it would be to get away from prints and syringes ASAP, learn agar, it makes you a better cultivator and it's flipping fun as hell (at least for me)
