Different agar medium
    #1817180 - 08/15/03 01:58 PM (20 years, 7 months ago)

hi there

Until now, I always made my agar medium following the PDA recipe
but then, I purchased some light malt extract and tried the MEA
none of my cultures look the same
oysters were used to be cottony
now the mycelium is so thin, I can almost see it
same goes for other mushrooms
now, I'm very sure I didn't mess up the recipe (I have a scale)

What could the problem be ?
the malt extract isn't good ? perhaps not enough ?
or is pda simply better?

another thing I can't figure out, what is neo-peptone ?
I heard it could improve the colonization speed
where could I buy it ?

Ya know. I have noticed that too. The colonies of typically heavy mycelial species (on my supplemented PDA) are wispy and even hard to see on the MEA that I have made. Further, I have found that spore germination is poorer and slower on MEA when compared to my supplemented PDA. I think it has something to do with nutrient levels. You might alleviate the problem by spiking the MEA with oatmeal, yeast extract, and peptone. I have not yet tried this myself though.

???

I know www.fungi.com sells sterile culture media but I'm not looking to buy mea or pda powders

It's the only place that sells peptone ?

I knew they sell it
why would I ask then "it's the ONLY place that sells peptone ?"
perhaps you didn't read my post carefully

does it really work ? it is for the hobbyist or the commercial grower ?
it seems pretty expensive for 100 grams

I think I'll go back to pda
perhaps supplement it with oatmeal

I'm sorry. I must have misunderstood.

I just checked. Sporeworks sells it too. Cheaper as well.

good luck!
