Oyster Mushrooms on agar
    #3124671 - 09/12/04 12:50 AM (19 years, 6 months ago)

I cloned a generic oyster mushroom from the supermarket on peroxide/agar/malt/yeast/rice flour/rabbit food/fir wood fuel pellet medium. Each petri dish is enclosed in a zip-lock bag they are being incubated in a styrofoam cooler with the lid on, which is inside a big chest sitting on one end that has a lightbulb connected to a thermostat in it, set at 75 degrees farenheit (very constant temperature inside cooler). 4 of the clones grew to some extent (1 contaminated with green mold, 2 turned into puffballs but the only growth was on the mushroom cutting itself) and only one actually seems to be spreading across the agar after about 2 weeks. Now my questions are should I take the dishes out of the zip-lock bags (was to prevent contamination) and should I be incubating oyster mushroom mycelium in a dark cooler like I am?

The dark cooler is ok. The best temperatures for incubation is normally about 78 degree Fahrenheit but 75 works very well, too.
If there is enough air in the ziplock bags it should be no problem but parafilm is better.

I like to get a roll of polyethylene food wrap and cut rings off it making minirolls i use to seal Petris

I really dont like parafilm.
Its a specialty buy, its opaque, it deteriorates to a sticky shitty mess

I prefer the transparent, cheap and available rolls instead.

I use polyethylene as well but I've never used parafilm. Works great, no complaints at all.

Have never tried polyethylene food wrap, so I cannot tell how it is.
But I think it is important that the culture has a bit of air exchange but does not dry out.

Thanks for that tip. I think I can get that brand here and it will be a lot cheaper than the parafilm.
