Colonizing petri dishes, what can i do with them now ?

Im looking for some suggestions or pointers to teks on what i can do with these petri dishes i have colonizing, I got a couple B+ and one menace strain doing well on some potato dextrose in petris, can i take this mycelium and make syringes from it, or anything like this ?

haha? i dont seriously think shrooms will grow right off the petri dish, will they ? i was wondering if i could take that mycelium and make syringes from it somehow to innnculate other projects

You don't have to use syringes to inoculate when you have pieces of actual myc on a petri dish.

Run a search. You can inoculate your jars (or whatever) using pieces of myc from your dishes.

The Mushroom Cultivator had some good stuff on petris.

Mind you, I'm just a noob.

Yes there are a few posts on doing it.  Drop a dime size of sterile water on the mycelium and rub it with you needle and then suck it up.  Otherwise just drop a piece of the mycelium in a jar of some grains or seeds.  Or make a LC with a piece of the mycelium.

yea I'd recommend making an LC and inoculating a Jar or two. Remember that the best myc to pick from the dish is the ropey variety since they are supposed to colonize better.

You can slice up a petri dish of agar and use it to inoculate at least ten quart rye jars.  After two weeks they'll be fully colonized, and you can do grain to grain transfers, turning each of those first ten into ten more.  In two more weeks they'll be fully colonized, and you'll have turned a petri dish into 100 colonized rye jars in 30 days.  Should you chose to repeat the grain to grain transfers again, you'll have 1,000 fully colonized quarts of rye grain in six weeks.  LC can't come close to that kind of performance.

RR: not to nit pick but when I g2g a jar into 10 others (usually 9 that's the max of my PC) it only takes about a week to fully colonize. Anyway I think an LC will give you a bulk quantity of a strain isolate to use later that's why I was suggesting it. Petri dishes make me nervous with all their open surface area and I bet this guy is using a glove box. A clean syringe full of LC is still a nice tool to get say his next batch up and going that's why I recommended both.

Yes and that makes it all the more strange that somebody would go to the bother, and have the knowhow to making petris, and yet not know what to do with them...

They can pin on agar.

lol :-) i knew you could spawn them to grain jars, i was just wondering if I could make syringes with them, why the hostility?!!? Im still a newb but im doing my best here!

mostly the reason im messing with petris is curiosity, plus trying to get experience with them for further in my career isolating strains and what not, plus primarily i was just using them to test the spore syringes i made from my prints to make sure they didnt get contaminated in the process, rather than making a whole jar and what not

It was answered, you can and if you do a search there is a ton of info.  Just use some sterile water on it and suck it up.  It is very easy.

Here is a post on it.  You can search "Agar Culture transfer to Liquid Culture Tek" to get the pictures.  But instead of making a LC just leave the mycelium in the syringe and use sterile water instead of malt.


You will need an agar plate with healthy mycelium growing on it. This example uses the Pearl Oyster.
You will need a sterile syringe and needle to make the transfer. Last but not least, you need a jar of liquid food. We are using a standard malt water mixture.



When working with agar plates you must work in a clean glovebox or in front of a HEPA hood. With this in mind we will continue.

To begin we will remove 2cc from the malt water mix.



Now carefully remove the lid on the dish. Locate the best looking mycelium on the plate, avoiding areas near the edge of the dish. Slowly squirt a small amount of the liquid from the syringe on this area. You want to be careful placing this solution on the plate. Your goal is to put a dime size ?drop? that will hold its shape from water tension. If you put too much on too fast it will run all over the dish, increasing the risk of contamination.



Now it is simply a matter of scraping the mycelium under the water ?drop? (with the tip of the needle) until some of it is suspended in the solution and then drawing this back in to the syringe. Working in front of a HEPA filter helps greatly here if you accidentally draw extra air in, but a good glove box will work fine also.



All that is left now to complete the transfer is to inject this syringe back in to the jar of malt water.



Your newly inoculated liquid culture jar should now be placed in an incubator at the correct temperature for the species. Once every 2 days you can swirl the jar around to re-mix the mycelium. Growth will be evident within hours with different mushroom?s mycelium. Growth will continue for some time but it will be optimal to use in 7 or 10 days.

I didn't mean to come across in a bad/hostile way (if that was referring to me), just thought it strange since petris are a sort of advanced step. I first did agar work in jars resting on their sides, by the time I got proper petris I had a few grows under my belt.

And well worthwhile. I personally have never had any luck with transferring colonised agar, or tissue to a LC, dunno why. I have good success with mulitspore LC.

thanks for all the feedback guys :-) i got a couple grows under my belt, couple being... 5 projects thus far that have fruited, couple in the works right now, i know this doesnt compare to the big guns out there but ive only been doing this 3 months i think, i think im doing well

thanks for asking snypster :-) well by the looks of it (though im not supposed to look, i peek in one of them) there is no dark black/brown poo left on top, only lots of mycelium, ariel and not, and where there is poo its light brown instead of dark, which i believe is because of the mycelium colonizing it, coming along well, i lift the lid one inch after spraying some lycol around and shine a flashlight in, and i take a whiff, smells great, familier myco smell with no hint of contams, and a light poo smell that i recognize thoughoghly by now, lol, im currently pasturizing a 5 gallon bucket worth in my oven, what work!!!!

i leeched it, dried it out on a tarp, shredded it in a blender (lots of work for 5 gallon bucket, lol, must find a better way!) rehydrated it to field capacity, mixed in ~20% field capacity vermiculite, stuck in 3 oven bags equally, and its currently pasturizing in my oven at ~175ish, lots of work, i just made up 8 quart jars of WBS as well, im pressure cooking those as we speek, lol, i have 6 pint jars of WBS already colonizing for about a week or so, when they are done im going to use 1 for grain to grain transfer, and hopefully 5 for this poo im pasturizing, dont know if that will be enough though, im going to make a monotub project with all this poo, in my big monotub, im gonna cover the holes i made and filled with polyfill with tyvek, and shotgun the hell out of it, dig ? hopefully 5 pints will be enough WBS to spawn to all this poo, or atleast enough of it to fill my monotub 3", what do you think?

the one to the right, old pic, im hoping to have 3" of poo in there, im going to do the math better tommorow, figure out how much poo i really got in these bags, and try to math it out how deep that will get me in that monotub, lol, not sure how im gonna pull that off, but at any rate how many jars do you think i will need to spawn to that monotub at 3"? if 5 pints isnt enough think it would suffice for 2" ? otherwise i will ditch the grain to grain transfer and just use the slices of mycelium growing on potato dextrose in my petri dishes to inoculate to the WBS jars im PCing right now, i only have 1 spore syring left (one i made mind you) it would prolly only suffice no inoculate half of these, i dunno lots to think about

I am guessing, but 5 pints sounds good. I'm not sure about the tyvek over the holes though. I would just stick with polyfil.

I'm the same way with my tubs. It is hard not to peak every once in a while. I have a few in progress right now. Should be ready to fruit tomorrow or the next day. They are only 5X8 trays, but they are deep. Straight coir. I'm not casing, just fruiting in a big shotgun with about 7" of perlite. I'm trying to keep my current project as simple as possible.
