So you need a glovebox or flowhood to work with agar?

I like working with agar because it's simple and hard to screw up but my recent experience has made it obvious that I can't open my jars to pop a piece of agar inside the jar. There must be so many spores floating around in this old house that even a few seconds of exposure is enough to contaminate the jars. Do most of you who work with agar use a flowhood or glove box?

it's not worth working with agar without a flowhood or glove box.

the cost of contaminated jars/cultures/plates can quickly equal, then exceed the price of a box or hood. put that money towards something good.

no flowhoods/gloveboxes are optional if you are like me,
that is a total isopropanole junky with boxers on his hair,
gloves and facemask and working at the speed of light.

if you can manage this, check out drooldonkeys peroxide agar tek,
which led me to this, entirely without glovebox/flowhood:

im sorry you are offended by my post as i seriously didnt mean to offend you.
please take a look at the picture of the dish showing aspergillus niger
to prove that i do get contams occassionally aswell.

btw, there is no need for you either to call me an idiot.

Yes you can work with agar without using a floowhood or a glovebox.

You would have to clean the living crap out of your work area though...and make sure that you don't have any drafts of air what so ever.

Your contam rate will be higher too.

I used to do agar work without using either...but I was extremely sterile in everything that I did.

I'd suggest building a cheap glove box...it will be a really good investment.

I've had little to no problems working with agar without either. However, I do use a hepa filter and work downstream from that.

That would still be like using a flowhood...just a cheap one.

True, Road. That is basically like a flow hood. Actually, I've never had any contams with agar (so far), and very few with the grain that I've transfered too. I'm having problems further down the line with casing, though... But that doesn't have much to do with this topic..

Thanks for the help guys..I've recovered from this morning's disappointment at finding all my jars contamed...gotta try again. I've spent too much money on pressure cooker, huge bags of rye, 15 or so petris, agar, HEPA filters, etc to give up on this.
I've probably spent about $700 CAD on this so far, which is quite a bit for me right now.

Now to construct a glove box..I hope this works!

now to be honest i have to start by stating that i do not do agar work. but in regards to the glovebox... here is a little tips that should work for that as well. "the clean bag", its something i came up with long ago, when i was thinking of writing a " cheap-ass, lo-tech" TEK for cultivation. I've used it plenty of times (when i still used to grow) for inoculations, making monocultures or anything that needs clean environs. anyway here it is.

-The Clean Bag-

the only things you need are:
-a (large) see-through plastic bag,

-one plastic/latex glove,
-spraybottle with disinfectant (h2o2).
-a bottle of sterilized water (optional).

sterilize all the things you will be working with and put them into the plasticbag (if you lay the bag on a table, you can easily arrange the things on the table, inside the bag).

wash hand, dry, put on glove. make sure there is some air in the bag. put the rubber bands on your gloved arm, and put your hand at the mouth of the bag. use rubber band to secure the mouth of the bag around your wrist.

pull the bag and rubberband a little to the side and insert nozzle of spraybottle, and spray everything inside the bag to sterilize it. use your gloved hand to make sure it gets everywhere. remove spraybottle (and if your paranoid use second rubberband to secure/double seal the bag around your wrist.

wait 20 minutes (read a book).

by leaning the table and placing your wrist lower than the edge of the table, you can easily drain out the disinfectant (avoid getting it on your naked wrist).

if you are going to work with spores (or anything sensitive to your disinfectant), this is a good time to use the optional waterbottle to rinse items and insides of bag. drain as before.

now. you should have a relatively clean working space inside the bag. and do whatever procedures you need to. i find it easiest to work with the gloved hand and hold items through the plastic bag with the outside hand.

when your finished, close all jars/dishes and just open the bag.


it doesn't get much cheaper, easier, and stealth than this.

EDIT: suitable h2o2 concentration is around 6-7%.

you want the total concentration of h2o2 within the agar not to exceed 1% of 0.3% h2o2.

yes, on or in stubstrate (like agar) you want lower ratios. but im not talking about actual agarwork, this is just about creating a sterile environ, forthe working.

but to sterilize the insides of the bag, outsides of jars etc. one needs higher conc. i of course assume that you have the sense to close all jars, sporesamples or whatever while spraying and transfering to/from bag.
