I'm not sure about there being more minor alkaloids in dried than fresh unless the fruits have been dried at low temperature. Tryptamine and 4-hydroxytryptamine degrade really easily and don't usually show up in HPLC results. And it's the addition of the phosphate group at the norbaeocystin step that stabilizes the synthesis molecule which is why norbaeocystin and baeocystin often show up in the results depending on drying method.

So as you say in one of your linked posts, the precursors aren't manufactured and stored so the minor alkaloid content in a fruit has to be the product of active psilocybin synthesis. And with the precursors being unstable logic dictates that there are going to be higher quantities of all the minor alkaloids in fresh fruits.

They've stopped presenting their results like this now but I spotted a pattern in MagicMyco HPLC results with regard to minor alkaloids and drying temperatures. Low temperature dried fruits retained the more stable minor alkaloids whereas elevated temperature dried fruits just had psilocybin and small amounts of psilocin. This is typical of the patterns of the alkaloid ratios between high and low temperature dried fruits.

The temperature isn't mentioned here but the alkaloid profile is typical of elevated temperature dried fruits in their HPLC results.



And two with low temperature drying alkaloid profiles with norbaeocystin and baeocystin remaining in the dried fruits.





You can't compare total potency as they're all different strains but it's interesting to note that the psilocybin content is higher in the elevated temperature dried fruits where only traces of precursors remain which may indicate that elevated temperatures increase the rate of psilocybin synthesis in harvested fruits until the enzymes are degraded over 50C and is the only evidence I've ever seen that might indicate that elevated temperature drying could be best for long term storage.

This hasn't been resolved by the research yet but due to the reactive nature of all the precursors and the chemically rich environment of the cytoplasm I think the psilocybin synthesis enzymes are likely to form a metabolon (they all link together) so the product of each synthesis step is passed directly to the next enzyme in the sequence to prevent it reacting with anything else. So the minor alkaloids likely come from the remains of the metabolon when ingested fresh or the fruits are dried or extracted.

Fresh fruits also have functioning enzymes that drive the reaction into psilocybin as you said yourself, the mushrooms are not storing the intermediates the way dried fruits do. The study that compared fresh to dried found much higher levels of all of the intermediates when compared to fresh (when dried at room temperature), obviously one can destroy and degrade them depending on harvesting, handling, drying method, storage conditions, ect but psilocin can be destroyed in similar ways. So if you want to experience the effects(or more accurately the lack there of) of all the minor alkaloids just dry them at room temperature :shrug:

We were talking about amounts not concentration. The concentration of all substances in magic mushrooms increases with drying as the fruits are 90% water and the amount of an unstable substance can go down as its concentration increases like it does with psilocybin during the drying process. 

What I'm saying is from examining HPLC test results it's clear that the amounts of the precursors and alkaloids contained in fresh fruits either decreases or they're completely destroyed by the drying process, especially at elevated temperatures, so logic dictates that like psilocybin there are higher amounts of all the precursors in fresh fruits.

Apart from that I think we're both singing from the same hymn sheet.

I agree that we are both singing from the same hymn sheet, I just think there’s a slight misunderstanding Im trying to clear up.

That study is talking about total content/amounts, they’ve already taken into account difference in water concentration if you take a look at it. The total amount of tryptamines do not change between fresh and dry, however the ratios and therefore total amounts of the specific tryptamines do. Mushrooms dried in the dark at room temp have far higher amounts of intermediates than fresh does, they directly compared the two from the same samples.

That logic is incorrect as the quote from the above paper shows; your appeared to be only looking at flawed drying techniques, dried in the dark at room temperature they do not get destroyed during drying, but their is a conversation taking place. That study shows there are higher amounts of all of the precursors in dried fruits, not in fresh..

I'm familiar with that quote from the paper and it's always stuck out to me as an anomalous result and an unsubstantiated statement. Every other paper that investigated potency and drying found that even room temperature drying led to a not unsubstantial loss of potency. Like this one.



And even in the "Stability of psilocybin and its four analogs" paper the quote comes from they found that potency degraded in dried fungal material at room temperature. ""Degradation of tryptamines was observed at 25C, 50C, 75C, 100C, 125C, and 150C for 30 min according to the procedure attached in the supporting information."



I'm pretty sure I've read all the published research on drying, temperature and potency and I'm not aware of any research papers that found that drying (apart from freeze drying) didn't result in a loss of potency.

I don't disagree with this statement and I've mentioned it previously. They're talking about concentration not comparing analyte yield between fresh and dried fruits. Baeocystin and psilocybin have increased stability compared to the other indoles due to the phosphate group which is added at the norbaeocystin to baeocystin step in the synthesis. Which is likely the reason they survive the drying process.

Higher amounts of the dephosphorylated synthesis indoles like tryptamine and 4-hydroxytryptamine are found in fresh fruits because they're unstable and degrade easily during the drying process or perhaps if psilocybin synthesis is still occurring after harvest they get depleted and/or the cofactors their enzymes need for their reactions run out. And the increase in psilocin in the fresh fruit results is likely to have occurred post harvest as it's now thought that there is no psilocin present in healthy undamaged growing fruits.

The Gotvaldová paper could be correct that room temperature drying doesn't result in a loss of potency on average as it's one of the most recent papers to be published on the subject but personally I doubt it, due the weight of the previous evidence including the same room temperature drying experiment that found that the drying process always results in a loss of potency.

That paper also shows that freeze dried has a higher amounts of alkaloids then fresh does so obviously somethings wrong there..

There was no change in overall tryptamine yield from fresh vs dry, so yes they are talking about both analyte yield as well as ratios, nothing is lost during drying at room temp in the dark, but they do change.. Losses can occur at room temp after drying when moisture is re-introduced like with exposure to humidity in the air as the enzymes responsible for degradation are stored outside of the cell wall.

This is correct, psilocin cannot exist in a healthy undamaged fungal cell because of PsiK, however how do you extract/consume fresh mushrooms without damaging the cells in the process? You can’t… That is why fresh almost always has a higher unconjugated psilocin content. It’s not because it’s being created independently in the cells, but that’s sort of irrelevant to consumption no?

don't know why you insist in using this table, lol. is obvious that is unreliable, the freeze drying one have three times more alkaloids than the fresh one.

you also seem to have changed your mind, as the same argument icy used against you, you used against me in the past.

i didn't read all the thread, but if what you are saying is that is hard to say that 100% of psilocin is converted back to psilocybin in dehydration, i actually agree. there's many factors to that, the main one in my opinion, being the inevitable fall of ATP, that makes psik enzyme useless.

so stuff like time of exposure, senescense, and light can make some differences in this absolute claim that nothing is lost. but i think that is fair to say that most is preserved, just again, not everything and every time.

That's the unsubstantiated statement I was talking about. All the other papers published on the subject recommend low temperature drying and processing to maximise the yield of tryptamines not because it stops them degrading completely. The Gotvaldová paper is the only published research to claim that there is no appreciable loss of potency during the drying process. Hence my scepticism.



The reason that freeze drying resulted in much higher potency than fresh material in the results of this chart is likely due to the fact that potency quickly degraded in the fresh sample during processing (as indicated by the increased psilocin content) and that fact that freeze drying ruptures all the hyphae and produces a highly porous brittle structure which when ground for sampling increases the efficiency of the extraction. Freeze drying also halts any enzymatic activity and all the other degradation reactions like hydrolysis and oxidisation so potency can't degrade in the process. So the most accurate figure for the potency of the sampled material is the freeze dried figure and the discrepancy is explained by the difficulty in getting accurate figures from fresh material.

They don't all survive though. Find me HPLC results that found tryptamine or 4-hydroxytryptamine in amounts comparable to the baeocystin and norbaeocystin content that's found in room temperature dried fruits. They're always notably absent as shown in the HPLC results I posted earlier in the thread.

The intriguing figures from that chart is the discrepancy showing higher potency in room temperature dried fruits compared to the fresh calculated as dry figure. That could be down to the inefficiency of testing fresh material as mentioned above or it be evidence that the fruits continue to manufacture psilocybin post harvest.
