Sounds like your agar is just a little soft. Not ideal for transfers, but once you're almost there it's really practical to make slurry / LI from. Nothing gets a jar started like inoculant dripping from the glass.

My cube transfer took and didn't contaminate, kinda surprised me. After all the effort I put in to trash that plate. Made another T1 two days ago.
Galindoi does nothing, also on BRF/verm.
The Gymnopilus print has been lost in transit for well over a month now.

Well, most of my pan cyan plates went bacterial.  my technique of going spore print to agar isn’t the most professional (I take a small bit of agar from the xacto cooling dish, rub it on spores, and then put into a sterile agar dish).  I could’ve stepped that up some, but live and learn.


One plate is showing a tiiiiny tiny bit of mycelium on it so I’m going to say my shroomy prayers and in 2-3 days hopefully take enough transfers to keep this train rolling

India Woodlover plates have yet to show any activity.  Keeping a close eye on them, though.  Only been like 4 days :smile:

Yup, think i got it, from ~10% rhizo to now 50+% :smile:

Very excited for these charcoal glass T3 plates, should be throwin straight ropes. Gotta control myself and let them cool until the morning lol.

Used 6g agar, 4g LME, 3g charcoal, ~300ml water

While my LAGM Neox spores didn't germ (which I re-streaked today) - the Stropharia alcis did.  The myc is a nightmare, and made me very suspicious at first.  It is radial on both germ dishes, and structured the same- which gives me hope.  It is more uniformly radial on T1s (example pictured against germ dishes) as well,  so there's that.  Fingers crossed.  Took some T2s today,  so we'll see if we go uphill or roll back down.  I notice that with a lot of random wild mushrooms you clone - the myc can be really random at times.  Even something like lion's mane or maitake tend to express irregular/non-uniform structures.  I hate using potato broth agar too - always gives weak ass growth which isn'thelping in this case :resignednonacceptance:
I'll be very happy if I manage to get wedges of this species to grain this month.

Pretty sure I’ve gotten duped and I think I need to change my agar recipe haha. Been running Milkboys pda and it’s been good to me for the most part.
But I’m seeing a trend that I think needs to be addressed.

So for some context:
This is an ochraceocentrata wedge that I received, you can see how the mycelium didn’t start to look fuzzy and white until it grew out substantially. I thought because it was no pour and super soft..


Initially, it looked just like these, which are crimini clones taken from the sterile inner stipe tissue a few weeks ago. You can see the typical form of mycelial growth.



And now, all the “contam” that my lagm plates have showed me..







Now, I typically simmer my agar to help incorporate all the dry ingredients before pc’ing.
Which I didn’t with this batch.
There were still chunks of unincorporated dry material visible.
Maybe it was too soft as well?

Gonna make some xfers with a new batch and see what happens.

Made 4 T1's of AA+.  All 3 plates had germinated.

Went swab > 3 pucks with a fresh PE swab, as there was no action on the 3 agar germ plates.

India Woodlover and Pan Cyan Peace River spores appear to have germinated!  Very excited :smile: 

will update when I take transfers in a few days.  I dunno if this pan cyan plate is clean but it isn’t shleppin bacteria like the other ones so that’s kinda sweet

I like flame sterilizing my #11 blade, place it on top of the closed receiving plate, wait a minute and then gently (using the blade tip) scrape off a few specks from the spore print into the plate in a manner that resembles a clam..the lid just half way open. Took me a lifetime to get to where i am now but for every decent grow i also had a million loses. Much love :heart::heart:

The cubensis T1s. The discoloration is soot from flaming the knife with a large candle, I know, that's not how it's supposed to go. Couldn't find the torch when the next day it was just standing on the table. Drugs r bad.
Galindoi is probably done for. This little pissdrop is all that grew out of my ancient spores. Important lesson: make fresh prints yearly. Learn freezer storage. I've grown this species since 2016, never used it more than say three times a year. It was however our absolute fav for dancing (high doses of Ps cyan are hit ir miss at crowded venues with 200 bpm music), but I fucked up on long term survivability.



A very kind member offered a swab of Gymnopilus, so my hopes of growing those are reinstated. I see you removed the offer from your post, so don't know if you want to be mentioned by name, but thanks for your generosity!

Grabbed a different Amazonian print, wet 4x swabs with sterilized distilled water, rolled them in the print, then broke them off and jammed into each puck. I also took the entire print and made a spore syringe with it. I used that syringe to also put some drops on the opposing side of each BRF puck.


I'm not convinced, but this might be some white fluff growing on this contaminated plate/swab.


Tico Rye jars are doing well. I did a break and shake on this jar after the picture was taken.


I'm fairly determined to get these spores to germinate, I'm going to see if I can find some rubber bands and glue the tip of the syringe shut to force hydrate it.

Jan 20 Update!

So, I see what strikes me as Germination in one of the remaining 2 GT plates! It took a bit, likely a mix of stubbornness and cold temps. But it's something! Not the best pic, but I can see a few patches, roughly circular. There are other competitors on that plate, though. So I'll likely xfer to a "just in case" BRF puck as well as a couple of fresh plates.



The other GT plate is very quiet, still. But it looks to have the least absolute growth. But it may just be shy/undetectable my monkey eyes atm. We'll have to wait and see.



As for the surviving P. Djamor plate...



It's looking grim. Looks even worse IRL :sad: Prolly gonna toss that, start the spores again into a puck. On the bright side, it'll make my log a little simpler to just keep track of GT. Specially due to some unrelated projects creeping in (myco and otherwise).

T1s will have to wait until Wednesday due to some work/paperwork/busywork stuff. By then, both GT plates should give me more information to work off of. Feeling nervous, but excited!

hell yeah congrats!
and are you trying to use a cell phone, a point and shoot camera, or a full size digital camera?

cellphone: download a camera app that lets you control focus manually. set it to a foot or two and move the plate back and forth until it's sharp. slightly over-expose as auto mode will try and even out the backlight and make the mycelium really dark.

point and shoot: not much can be done, enable macro mode if it has one.

full size: manual focus w/ macro lens, set the exposure to 'single point' on the agar and blast away. Preferably in RAW format.

I've been using a cell phone. The main issue is being able to see anything with backlighting, but a friend is coming by later to help me get it sorted.
