You right i transfer way to many half a Petri Dish in small pieces and when you say it it's clear that's not good i try too makes thinks quicker end end up with more time  I let this Jar running and make a new smaller Jars with Grain and transfer again after colonized i transfer G2G to a Bigger Jar that's what i will do with less Agar.. Hoe many Pieces?

Okay, working with a strong language barrier so I will be blunt.

You do not have the skill to do grain to grain transfer.  This grain is not clean and I would not even use it to top fruit.  It is time to start over at the agar level.

One wedge of agar per jar, make it about the size of your scalpel blade.  Only select from the leading edge of a clean culture.

Thank you so much. I was growing concerned as everything I'd read to this point says about 2 weeks for this step.

So when I've got a shoebox like that that's fully blanketed with fruits. do I still do a twist and  pull for Harvest or is this an instance that may better to float it and use a scapal  or knife or something?

Personal preference on that one.  Everyone's got a reason why their harvest way is the best but far as I can tell

So I have a pair of sterilizing sleeves that I put into my pressure cooker and I realize now I didn’t cover the holes with anything.

Now I’m gonna just open it in front of my filter fan unit. Should I have just covered them with some poretape or something?

I’ve been growing from an AIO bag and seem to have had pretty good harvests. I guess my concern is the length of the stems.  Some look “normal” some are super long and fat. Some long and skinny.  I tried to get a pic of some different shit to show the size difference.

Could be reaching for light?  Maybe not enough FAE?

Probably an FAE thing.
Considering it was a bag, you could try opening it a little more next time or maybe poking another hole in it.

Yes. Medical grade straight form the roll .it's as close to sterile as you can add.

So you took your conjecture and warped it into fact which is exactly what I said must stop on the shroomery.

Not trying to be harsh but there's just so much confirmation bias here it becomes a bit counterintuitive.

In my experience, there are 3 big factors in PF cake colonization times: what you inject in them (spores vs LI/LC), amount injected and ambient temperature. If you use a spore syringe it will take at least 1-2 weeks longer than using an agar plate as Liquid Inoculant. I also tried experimenting with 1cc per cake vs 4cc and the 4cc ones colonized faster (I used less water in that brf mix to balance it out). And also in summer the growth was just faster.

Hi.

I've decided to try grow APE.  I got told it is "hard" to grow, and "slow".  Any truth to this?  I'm up for a challenge

Also, is there a dedicated thread on this site that I can go study?

Ciao, and TIA

It's just as easy as any cube , tad bit slower to fruit but stick to the plan and you'll be fine

cool thank you. ive been lurking around here since the lava rock days but only recently started getting into agar and making my own prints. trying to wrap my head around the whole generations thing. F1 F2 etc.

if i get a print from a vendor, say Ecuador, isnt that technically like an F347,842,201? im wondering why start at F1 when i take a print from a "desirable" fruit and grow it out. does it really mean anything or is it just a point of reference??

i guess thats not a quick question but if anyone has a link to in depth info on the topic, id greatly appreciate it.

Yep most old normal varieties are pretty inbred so nothing wrong with a ms grow , far easier to get a clean culture than look for a clone afterwards

As for starting at F1...you really don't. The only time you're truly starting at F1 is when you introduce new genetics via crossing.

ahh i see. thanks!

that said, ive been working with PE6 for a while and did swabs of them to agar for the first time. ive got a handful of totes made from those different plates, each exhibiting their own distinct characteristics. wondering if there are  benefits to cloning from these totes or if i should just take more prints/swabs to try to "isolate" these characteristics...
