Inculcating agar with a print or syringe?

What is the best way to inoculate Agar, spore print or a syringe??

Are there pros and cons to either?

Personally I like a print. Syringes are good for pf Tek but for agar it's just an extra step

Both work great.

No cons really for either.  It's all preference really.  Some people say syringe to agar is risky but it has always worked as good as print to agar for me.

you could, yeah that's how some folks do it.  Honestly, my method is just to cool down my scalpel in agar, get a small piece of it, rub it on the inside of the print somewhere, then put it back in the agar.  I've heard of folks using loops and doing a Z streak and it working great tho.. same idea as my way basically.  I mainly just recommend making sure you have a little agar on your scalpel/loop to catch the spores with.  You got the idea though!

My preference is prints as well...but either will be fine.

If you only use a small portion of the print you'll be cutting down on any possible contaminates germinating along side the spores(if the print is dirty)

If you use a syringe and the print is dirty you'll end up with more bacteria(not common) or mold spores(common) making it more difficult to isolate out a clean wedge.

If your syringes are commonly contaminated with bacteria then either find another sponsor since their source of spores would be the problem but most vendors run quality control checks on their supply's prior to shipments. Or if its homemade syringes your talking about.... then, did you sterilize the syringe prior to adding spores? And was the print used from a reliable source?

I can send you one if you want, guaranteed contaminate free. Straight from the bag to your sterilized syringe with sterilized water, impossible to fuck up. I use to donate to fsrc and e as well as give prints away by the masses to newbies...I'd hate myself if I sent out dirty prints but no one ever complained.....must have done something right.

Put him on ignore. :shrug:  I like prints as they’re readily available through your own work or trade.  If you’re working agar, there’s no real reason to make syringes in my mind.  But, if you only have syringes available to you for some reason, there’s no better way to get good cultures from them than by putting them to agar.
