how thick do you pour your plates?
    #2590657 - 04/22/04 10:42 AM (19 years, 11 months ago)

Im not asking to know how much to use, but i know ive use the reccomended amount and it worked fine. Ive also thought why use so much agar when the myc only grows on top and poured just enough to cover the plate (1/16") and i didnt notice much of a diffrence bedised cuting wedges its a bit thiner so a bit harder, but not much. This can save you allot of agar.

Thick agar holds more moisture and doesn't dry out so quickly. That is important when you grow slow species that take more then 10 days to grow over entire petri or when you don't plan to use the agar plate immediately.

Thin agar sticks to the sides of jars.

Here is a nice way to eliminate the condensation.

I agree with zeronio on this one. I often prepare many plates to use at a later date and certain species are slow to colonize agar.

As daywalker pointed out, Anno's agar tek works well at reducing condensation, just make sure not to knock over your cup of coffee!  I have had contamination on the uppermost plate with peroxidated agar before though, perhaps the heat is too intense and breaks down the H202?
