Alternatives to Agar Plates?

Hey Shroomers,
I hope this fits well in Advanced Cultivation -I also looked through the FAQ's & did a search for this so,
Here's the deal, My agar plates are history. -But we still have plenty of P.D.A. & yeast left.
Anyone ever try using 1/2pint or 1pint jars instead of the damn petri's? -Or are there any suitable alternatives besides the petri's? --Like from the grocerty store or whatnot.
Could anyone solve my dilema without having to order new petri plates.?
Thanks, Keep shroomin, GG

You have seen a tech on the net but cannot remember.. -Anyone else remember???
-Oh goodie, i think i should be able to figure this out then..
But i wonder which one would work better, The 1/2pint or the 1pints..
..also, i wonder if i could pour agar into each jar and then steralize for the 20 minutes --This way would bypass the pouring of the hot liquid..??
Any other input on this subject would be nice, The lab technicians would like to do this project this weekend.
thanks,

iv tride soap boxs i got at a pound shop then clean with alchole not great try clean the plates with alchole after a wash or two with dilute bleach the trick is to get a stirle place to let them dry jars work fine though but 20 plates cost me E3.50 so i dont bother and always get 95% contam free from stirle print and some wat less from wild strains washing and drying takes too long and are only about 60-70% cotam free

1/2 pints are easier to work with, but deep enough that you can sterilize the agar "in place" without any hassle.
That about all the tek you need!

Excellent,
By saying "in place" - i assume that you mean you can pour the unsterilzed agar mixture into the 1/2pints Before sterilizeing them in the pressure cooker??

yah, just use them like agar plates. Why do you need a special tek?

Hell yeah, this idea sounds great!!! Being able to seal the jar by tightening down the lid would be much easier and effective in keeping out contaminants, as opposed to using plastic wrap or parafin right?? And what would be better, lifting the lid to inncoculate or poking a small hole in the lid and covering w. tape afterwards?? Am I correct in my assumptions?? Thanks everyone!!!

If your going to do "clones" from fresh mushroom material, rizo material off anything, or wedges from another plate? Your going to have to open 1/2 pints, to do so.
But, no big deal -- IF -- you have a clean way to do it (clean room, glove box or flow hood).
Hell, a foaf started agar use with baby food jars & that worked fine.

You got it! One less chance for contamination.Though if you are doing cultures as oppsed to spores then I highly recommend peroxide agar.

If you can make Agar then you should really try pint jars...
Make as usual and PC.. Then twirl the jars around and spread the agar up the sides of the jar as far as possible... An uncontaminated jar will grow mycellium up and around it all over, the stuff looks like its taking over... Throw an uncontaminated penny in the jar and fill with water. Shake vigorusly then extract liquid with syringe and youve now got syringes that work better than and spore syringe.
I will never use plates again..

-Well i did 5 Pint jars today, but i did not twirl the jars around like you suggested. Seems like a great fu*kin idea though dude!!!
I will do the "twirl" next time. lol

One reason 1/2 pts are better than baby food jars,IMO,is that it is recommended to use wedges from close to the center of the agar before mycelium gets to the sides of the jar. With baby food jars, that doesn't give you much latitude. Also, canning jars seal easier and cleaner than commercial jars...

Well the lab tech's did the 5 one pint jars And i am rather happy with this.
So i instructed them to crank out the old HEPA,
And innoculate the following; 3 with Psilocybe weilii & 2 with Panaeolus castaneifolius
The technicians will keep me posted & I will do the same for you all.
Thanks again,

Yeah, I did some jars last night of agar. They dried and look good. Today I scraped some spores onto two of em. So we'll see how it goes. I''ll post an update later on!!

Two questions:
1. Is it ok to store already poured plates of agar in the fridge until ready to use?
2. Is it ok to turn the plates upside down so that condensation will not drip and land on the agar?
Thanks!!!!

Let me take a stab at this one guys,
Visigoth,
1. Is it ok to store already poured plates of agar in the fridge until ready to use? -From what i have learned, the fridge might be the most contaminated zone of your house. I would not store petri's in the fridge unless they were all wrapped, sealed and put into a contaminant-free container..
2. Is it ok to turn the plates upside down so that condensation will not drip and land on the agar? -No need for this. The condensation does no harm..
?? Correct/Expand on any of my answers if needed..
