Sterilizing Glass Petri FAIL * 1

This is my third time using my new glass Petris.  The first two times I oven sterilized and it worked well.  This time I had room in the pc so decided to foil wrap 6 per pack and put in with my grains.  All 6 are showing contams.
I also did some on plastic Petris and everyone of them are good.

How does everyone sterilize their Petris?

How much did glass petries cost you? I thought about trying glass petries for no pour containers but ended up doing no pours with MEA in 125mL jars. Sterilized in PC. Is working well, minimal contams.

I don’t remember exactly but think a dozen was around $60. 

Thought I would use plastic 60mm for transfers then grow out on glass 100mm.  My compromise to cut waste but when you have to oven sterilize for two hours or more it’s still wasteful, just in a different way.

I have around 50 glass petris that I dont use anymore. No pour is the shit, fuck agar, too, I do straight to 125ml baby food jars using brf+verm pucks...


It feels liberating to actually nail this isolation game.


in conclusion, I suck at agar. hahaha

It just seems weird to me that 2 hours in the pc didn’t work.  Or was it when I brought it out to the SAB?

You're assuming the problem was the Petris. It's probably your media container for the agar.

I was gonna say it was probbably contam that entered when you were pouring the agar, but TC knows better.
It also could be something you do during the pc cycle (unlikely though)

Oh, that’s definitely possible.  I had the other plates poured already from a different media bottle. It’s jogging my memory now, when I reheated the agar, in a pot of water on the stove, I forgot to loosen the cap (slightly)and it built pressure inside.  There was a sound when I opened it that made me cringe a bit. 

That renews my hope, going to try the dishes again.

Glass dishes are not for no pour.

You sterilize the glass dishes empty, then pour them like you would with
disposable dishes. If you sterilize them with media like no pours, then they
boil out and suck comtams into the dishes on cool down.

How did you sterilize them?

I pour agar into glass petris before PC'ing and have no issues with boil over or contamination.

I have oven sterilized the EMPTY dishes but am always a bit confused on temperature and time.  I’ll go 2 hours at 375 but not confident that is right.  The last time though I tried to sterilize EMPTY (wrapped in foil) in the pc, 18lbs for 2 hours along with my oat jars. Had it on top rack.

I have only ever done poured agar.  I will make media bottles of agar ahead, pour some extra plates and and reheat pour the rest as needed.  I do also make sure to use the whole bottle and if I don’t I’ll dump the rest.  I don’t cap and reuse out of the same bottle.

I'm pretty new to all this a couple months ive been playing with agar and when i jumped the gun i bought 24 60mm glass petri's I've been pressure cooking them along with my media bottle of agar for 45min @ 15 ps1 in these pp5 containers stacked upside down, i leave the lid loose on the container and leave them upright in the pc, when i unload the pc i tighten the lid and invert them into my SAB for pouring, i use the container lid as a stand and the container as a sleeve, i pour and return the container to let cool. 8 fit in the large and 4 in the small nice and snug.



Of course they have gotton a soap and water bath after use as well.

10m at 15psi would sterilize glass dishes. Getting them out without compromising them is where you most likely fucked up

I've been using the same glass petris no-pour for years. About a dollar each. No special process. Mix the agar in a small pot, pour into dishes, wrap 5 in foil, PC for 30. Never had a problem.
