You can get lucky, and some traits may be more consistent than others... thing about growing a bunch of mushrooms looking for the perfect isolate, is that you've got A BUNCH OF FUCKING MUSHROOMS!

You make a good point

Transfer the outermost circle to agar. Wedges are for grain inoculation. There are parts of the outer circle that are mostly isolated and mostly variated. Grab some good looking stuff with a tiny tiny half grain sized bit to lower variation. Agar biopsy with a needle would do great as well.

You've done your homework and I appreciate your responses.  That said, I don't think I've made my goal clear based on the feedback.  My goal will be to select a fruit, take a section to agar for cloning, and using the agar in noc'ing up some grains.  The catch is instead of using wedges for isolates I want the whole series of strains (since they did well together the first time).  Easiest way I can think of to do this is cut out a full ring of the agar (think of the agar dish as a dartboard with the fruit flesh in the center).

When you clone a fruit and different sectors appear on agar, were all those sectors expressed in the fruit? Or only one of them, while the fruit carried the rest of the genetics that we can now see on agar? Apologies if this has already been answered in this thread but I don’t quite understand.

Each sector will be a different strain, but may very well be indistinguishable subjectively... I would think (speculation) that fruit exhibiting recessive traits, would be fairly consistent at displaying it, simply bc it was present well enough to display in the pheno... I could be way left feild though lol
