non-agar gel matrix cloning in a pitri dish?
    #2519978 - 04/02/04 09:21 PM (12 years, 8 months ago)

So... taking stock of my cloning/ strain isolation supplies after a liason from said activities, and i realized i've misplaced my agar. I look at this as an opportunity to develop a new way of doing isolations. back in the undergrad days, i did some protein electrophoresis work using potato starch gels, which were made with 100% potato starch in a buffer solution. they solidified just like agar does, but at slightly lower temperatures. so, i was wondering if anyone has tried to do pitri dish work in a gel matrix without using agar (using starch or gelatin), or, alternatively, if anyone knows of any good sources for such work offhand. I discovered that several people have done gelatin work, but couldn't find any actual teks used.

This site is for teachers wishing to introduce their students to the world of fungi without outlaying large amounts of money.

They have a few recipes for gelatin... Don't know if this is useful or not.

I was under the impression that gelatin broke down at autoclave temperatures, thus was unsuitable for sterile work. Wrong?

I have been wondering for a while if gelatin could not be used but with a highter solids content such as in jello jigglers . The gummy worms and bears do not break down at very warm conditions . The only reason that i could see that would not work was maybe not enough water content . I am sure interested if someone does some work on this , in fact with this information that sakura has guided us to I have new hope on my ideas.
