If you reaaalllly want to mess with the contam cake you could (but really only in a very clean environment IE: your flowhood). Clone from the inside of the cake in front of your hood. You could take a sharp, clean knife and cut a wedge type section from in the cake, use just the innermost piece to clone. Use the least contaminated piece you can find that has not been exposed to the outer world (nasty spores.)

Use glad wrap to keep the containers well sealed and it will be fine with the hood off. You can also put them in the fridge if you want to.

Better not to mess with the contaminated cake unless you have to, but yeah -- no reason you can't. Just make double sure you don't allow contaminant spores to be floating around in what's supposed to be your sterile environment, otherwise it may contaminate lots of things. If the plates don't have antibiotics in them it would probably be better to put them in a clean baggie in the fridge (as cow shit collector said above) and work with them in a clean environment.

Spore syringes don't really have that many spores per drop in them, so it usually takes a while if inoculating from the actual spore syringe and very few points of germination. Better to use a spore print.

You could clone the inside of the cake, or a freshly picked fruitbody -- just wipe the outside with alcohol, cut it with a sterile blade and take a section of the inside to clone. This should be done in a glove box or flow hood, if you can.

Update- plates sat over night in clean room. They were innoculated the following day with syringe. And yah my friend is a little worried about the "spore count" going onto the agar. Is it a problem if he couldn't physically see the larger black clumps on the agar? He injected 5+ drops into each. Approx how long untill signs of growth? Well hopefully one will work for him, thats all the needs really, he is going to grain after colonization. Thanks again.

Depends on how potent the spore syringe was. You'll see growth eventually, though -- just keep them free from contaminants.

make sure you shake syringes well before innoculating agar also, espically with such a small ammount of innoculant.
