Flipping agar over to seperate mycellium from bac. contam
    #2247321 - 01/16/04 10:21 PM (20 years, 2 months ago)

I have accidently seperated contamination from mycellium by banging on the petri dish while its upside down until the agar flips over onto the cover. The mycellium has to grow through the agar, and sheds the contamination. This seems like an easier method than pouring agar over the mycelium and risking burning it to death. The only problem is that the agar is sometimes hard to flip.

Are there any oils, or anything else I can wipe the bottom of the petri with that would reduce the affinity of the agar to the plastic, but still allow mycellium to grow through it normally?

YOU CAN FLIP IT OVER!!!!!!!! OMG! i never thought about that...some of my prized wild prints came out with a bacterial sludge on them, and i tossed them...would flipping them have caused the mycelium to grow up through the agar and leave the bacteria on the other side to get a good clean culture???!

I've seen that trick before somewhere but I can't remember where.

there are considerations with this technique, other than the agar being hard to flip.

1. when you are banging on the dish, you are spreading contaminants. this is especially true with mold spores.

2. the environment inside the dish is exposed to the contaminants for a period of time.

when doing this, it would probably be better to flip the agar asap and as carefully as possible.

This trick is described in the Peroxide manual.
